摘  要:文章利用重组新城疫病毒(rNDA)治疗肝癌肺转移鼠,将免疫调节基因白细胞介素2(IL2)插入rNDV基因组,拯救获得rNDV-IL2。MTT法检验rNDV-IL2对肝癌HepG2细胞体外抑制效果。通过小鼠尾静脉注射H22肝癌细胞,构建肝癌细胞肺转移模型,腹腔注射rNDV和rNDV-IL2,治疗肝癌肺转移模型小鼠。结果表明,rNDV-IL2有效感染HepG2细胞,表达外源基因IL2并抑制HepG2细胞增殖。重组新城疫病毒rNDV-IL2治疗试验动物后,体内IL2基因表达量提高,重组新城疫病毒有效控制转移灶形成,且显著促进T细胞增殖,提高试验动物存活率,未治疗组、rDNV载体治疗组和rNDV-IL2治疗组30d成活率分别为0、50%和77.8%。In this study, the recombinant newcastle disease virus (rNDA) was used to treat lung metastasis of hepatocellular carcinoma. The immunoregulatory gene interleukin 2 (IL2) was inserted into the genome of NDV to rescue rNDV-IL2. Inhibition of rNDV-IL2 on hepatocellular carcinoma HepG2 cells were examined by MTT assay in vitro. Hepatocellular carcinoma cell lung metastasis model was established by injecting H22 hepatoma cells into the tail vein of mice, and rNDV and rNDV- IL2 were injected intraperitoneally to treat the metastasis model mice. The results showed that rNDV- IL2 effectively infected HepG2 cells, expressed the exogenous IL2 and inhibited the proliferation of HepG2 cells. rNDV- IL2 treated animals expressed the increased IL2, effectively controlled the formation of metastases, significantly promoted the proliferation of T cells and improved the survival rate of experimental animals. The 30- day survival rates of the untreated mice, DNV vector treated mice and rNDV-IL2 treated mice were 0, 50% and 77.8%, respectively.
