摘 要:目的研究新疆芹菜根提取物(AGRE)改善体外HepG2细胞脂肪堆积的作用。方法建立游离脂肪酸(FFA)诱导的HepG2细胞体外脂肪堆积模型,CCK-8法筛选AGRE对HepG2细胞的安全浓度,设立空白对照组(MEM完全培养基)、模型组(l.5 mmol/L FFA)、菲诺贝特组(l.5 mmol/L FFA+100μmol/L菲诺贝特)、AGRE组(1.5 mmol/L FFA+330μg/mL,1.5 mmol/L FFA+430μg/mL AGRE),各组细胞给药干预24 h后,油红O染色,观察各组HepG2细胞形态。按试剂盒操作方法测定甘油三酯(TG)、总胆固醇(T-CHO)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)等生化指标。结果与对照组比较,AGRE组和菲诺贝特组HepG2细胞中脂滴数量明显减少,HepG2细胞脂质相互融合度降低,TG和T-CHO的含量降低,LDH和SOD活性增高,差异有统计学意义(P<0.05)。结论 AGRE具有良好的体外调脂活性,可保护肝细胞、抑制脂质堆积,对HLP有较好的防治效果。Objective To investigate the effect of the extract of Apium graveolens L. root(AGRE)in Xinjiang on scavenging the fat accumulation of HepG2 liver cells, and to explore its effect on the prevention of hyperlipemia(HLP). Methods The hepatic steatosis model of HepG2 cells was established by treatment with free fatty acids(FFA). The safe concentration of AGRE in the HepG2 cells was determined by CCK-8 method. Cells were randomly divided into five groups:control groups(MEM culture media), model groups(1.5 mmol/L FFA), fenofibrate groups(1.5 mmol/L FFA+100 μmol/LFF), AGRE groups(1.5 mmol/L FFA+330 μmol/mL, 1.5 mmol/L FFA+430 μmol/mL). Each group was administered for 24 h and the lipid droplets were stained with oil red. The morphology of HepG2 cells were observed under an inverted microscope. Biochemical parameters were determined using commercial triglyceride(TG), total cholesterol(T-CHO), lactated dehydrogenase(LDH), and superoxide dismutase(SOD) kits. Results Compared with control groups, the number of lipid droplets was significantly reduced and the phenomenon of lipid interfusion was alleviated in FF groups and AGRE groups. The contents of TG and T-CHO in AGRE groups were decreased(P<0.05), and the activities of LDH and SOD were increased(P<0.05). Conclusion AGRE has good lipid-regulating activity in vitro;which can protect hepatocytes from lipid accumulation. It has better preventive effect on HLP.
