摘 要:目的探讨枳葛口服液含药血清对乙醇诱导L-02肝细胞氧化损伤的抗氧化作用及其机制。方法将接种于6孔板上的L-02肝细胞分为6组:对照组(10%正常大鼠血清培养)、模型组(2.5%乙醇处理)及4组不同浓度的含药血清组(2.5%乙醇+不同浓度枳葛口服液含药血清),24 h后测定各组细胞活力及细胞内氧化应激指标及抗氧化酶活力变化。结果与对照组比较,模型组的细胞活力、氧化应激指标丙二醛(MDA)、活性氧自由基(ROS)及抗氧化酶超氧化物歧化酶(SOD)等指标差异均有统计学意义(P <0.05);与模型组比较,各枳葛口服液含药血清组的以上指标均有所逆转改善,且改善程度与枳葛口服液含药血清浓度呈负相关,即浓度最小组逆转效果最佳,且与对照组比较差异无统计学意义(P> 0.05)。结论一定浓度的枳葛口服液含药血清能显著降低L-02肝细胞中氧化应激指标MDA、ROS含量,增加胞内抗氧化酶SOD活力,从而减轻或抑制乙醇诱导的氧化应激损伤。Objective To explore the antioxidant effect of serum containing Zhige oral solution on ethanol induced oxidative injury in L-02 hepatocytes and the mechanism. Methods L-02 hepatocytes seeded on 6 well plates were divided into six groups, one control group (cultured in 10% normal rat serum), one model group (with 2.5% ethanol treatment) and four groups of serum containing drugs at different concentrations (2.5% ethanol + serum containing Zhige oral solution treatment at different concentrations). After L-02 hepatocytes of all groups were cultured for 24 hours , the changes of the cell viability , the intracellular oxidative stress index levels and antioxidant enzyme of all groups were measured. Results Compared with the control group, the cell viability and the levels of oxidative stress index , like the activity of malondialdehyde ( MDA ), reactive oxygen species ( ROS ), superoxide dismutase ( SOD ) in the model group changed significantly (P < 0.05 ). Compared with all the tested indexes mentioned above in the model group , the indexes in the four groups of drug - containing serum showed varying degrees of reversal, and the degree of reversal was negatively correlated with the concentration of Zhige oral solution , that was the minimum concentration group had the optimal reversal effect. There was no significant difference between the minimum concentration group and the control group (P> 0.05 ). Conclusion Serum containing a certain dose of Zhige oral solution can significantly reduce the content of MDA and ROS in L-02 hepatocytes and increase the activity of SOD , therefore alleviating ethanol-induced oxidative stress damage.
