摘 要:鸡γ干扰素(ChIFN-γ)是一类具有抗病毒和免疫保护等广泛生物学活性的禽病防治有效药物,通过植物生物反应器表达生产是降低其生产成本和扩大临床应用范围的重要途径。为获得可以利用的ChIFN-γ转基因烟草生物反应器,本文在对T1代转ChIFN-γ基因烟草(TP)进行遗传稳定性检测的基础上,对其株高、叶片、根系及活力等指标进行测定,探讨外源基因表达对受体植物生长发育的影响。结果表明,TP烟草幼苗在移栽后植株较高,生长较快,叶片较多。株高增长以第7d时最为明显,3个株系的相对生长速度分别为野生型(WT)的253.23%、280.12%和260.42%;TP-1、TP-2和TP-3株系叶片数相对生长速度分别在第28d、28d和14d时达到最大,为WT的117.33%、118.84%和143.79%;在15d时TP株系组培苗的生根数和根系长分别为WT的9.09倍、7.06倍、5.06倍和2.23倍、2.66倍、2.41倍;水培条件下TP株系具有较高的根系活力,以第10d时最明显,分别为WT的1.12、1.44、1.39倍。因此,转ChIFN-γ基因烟草具有一定的生长优势,未表现出生长抑制现象,研究结果为ChIFN-γ转基因植物生物反应器的进一步应用研究提供了依据。Chicken interferon-gamma ( ChIFN-γ) is an effective therapeutic for poultry disease prevention and treatment with broad biological activities such as antiviral and immune protection. Through plant bioreactor expression of ChIFN-γ gene is an important way to reduce its production cost and expand its clinical application range. In order to obtain a usable bioreactor in ChIFN-γ transgenic tobacco, based on the genetic stability test of T1 generation of ChIFN-γ transgenic tobacco (TP), the height, leaf, root and vigor of the plants were measured to examine the influence of exogenous gene expression on the growth and development of recipient plants. The results showed that TP tobacco seedlings had higher plant height, faster growth rate and more leaves after transplanting. The increase of plant growth rate is most obvious on the 7th day. The relative growth rates of the three TP tobacco lines TP-1, TP-2 and TP - 3 were 253.23 %, 280.12 % and 260.42 % times of the wild type (WT), respectively. The relative growth rate of leaves reached the maximum at 28 d, 28 d and 14 d for TP - 1, TP - 2 and TP - 3 tobacco lines, respectively, and were 117.33 %, 118.84 % and 143.79 % times of of the WT. At 15 d, the number of roots for the tissue culture seedlings of the three TP tobacco lines were 9.09, 7.06 and 5.06 times of the WT, respectively, and the root length was 2.23, 2.66 and 2.41 times of the WT, respectively. TP tobacco had higher root activity under hydroponic conditions, which was most obvious on the 10th day, being 1.12, 1.44 and 1.39 times of the WT, respectively. The study indicated that ChIFN-γ transgenic tobacco certainly had growth advantages and showed no growth inhibition. The results provide a basis for the further application of ChIFN-γ transgenic plant bioreactor.
