桂枝茯苓丸加味对子宫肌瘤模型大鼠的影响

摘　　要：目的研究桂枝茯苓丸加味对雌孕激素负荷法建立的子宫肌瘤大鼠模型的作用机制。方法 将大鼠随机分为正常对照组、模型组、西药组、中药大剂量组、中药小剂量组及中药预防给药组。采用雌孕激素负荷法建立子宫肌瘤大鼠模型,观察桂枝茯苓丸加味对模型大鼠子宫组织形态学、血清及子宫雌激素受体(estrogen receptor,ER)和孕激素受体(progesterone receptor,PR)表达的影响,从而探讨桂枝茯苓丸加味治疗子宫肌瘤的作用机制。结果光镜下模型组大鼠子宫组织呈增生象改变。与模型组相比,中药大剂量组、中药小剂量组、中药预防给药组、西药组子宫湿重、子宫分角下直径均明显减小(P<0.05);中药小剂量组、中药预防给药组分角最大横径也明显减小(P<0.05)。中药小剂量组、中药预防给药组血清ER水平明显下降(P<0.05);中药大剂量组、中药小剂量组、中药预防给药组子宫组织ER表达均显著下降(P<0.05);中药小剂量组、中药预防给药组子宫组织PR表达均显著下降(P<0.05)。结论 桂枝茯苓丸加味通过减少ER、PR的表达,以降低子宫组织对雌孕激素的敏感性,从而达到拮抗子宫肌瘤增生,治疗子宫肌瘤的目的。Objective To study the intervention and mechanism of Guizhi Fuling pill on the uterine fibroid model of rat established by estrogen and progesterone load method.Method The rats were randomly divided into groups,namely,the normal control group,the model group,the western medicine group,the high dose treatment group by Chinese medicine,the low dose treatment group by Chinese medicine and preventive administration group by Chinese medicine.The uterine fibroid model of rats was established with estrogen and progesterone load method and the effect of Guizhi Fuling pills on morphological changes of uterus,serum and uterus estrogen receptor (ER) and progesterone receptor (PR) expression were observed in order to explore the intervention mechanism of Guizhi Fuling pill in the treatment of uterine fibroids.Results Under light microscope,the uterine tissue in the model group showed hyperplastic changes.Compared with the model group,the uterine wet weight and the diameter of left uterine horn in high dose treatment group of Chinese medicine,the low dose treatment group of Chinese medicine,preventive administration group as well as the western medicine group were all significantly reduced (P <0.05);maximum diameter of uterine horns was significantly reduced in the low dose treatment group of Chinese medicine and preventive group of Chinese medicine (P <0.05).The ER level in serum was significantly reduced in the low dose treatment group of Chinese meidicne and preventive administration group of Chinese medicine (P <0.05).ER expression in uterus tissue was significantly reduced in high dose treatment group of Chinese medicine,the low dose treatment group of Chinese medicine and preventive administration group of Chinese medicine(P <0.05).PR expression in uterus tissue was significantly reduced in low dose treatment group of Chinese medicine and preventive treatment group of Chinese medicine.(P <0.05).Conclusion Guizhi Fuling pill by decreasing the ER and PR expression reduces the sensitivity of uterine tissue to estrogen and
