梅花鹿瘤胃液中芽孢杆菌的分离鉴定与系统发育树构建

摘  要:梅花鹿瘤胃液85℃水浴20 min,接种于普通营养琼脂培养基,挑取菌落,经纯化培养后观察其个体和菌落形态特征,对分离菌株进行生理生化性状鉴定,最后通过对细菌16S r RNA测序,建立系统发育树。结果表明,7株分离菌中2株与枯草芽孢杆菌的序列相似度在99.93%以上,1株与地衣芽胞杆菌的序列同源性达99.76%,3株与赖氨酸芽孢杆菌的序列相似度在99.58%,1株与解木糖赖氨酸芽孢杆菌的序列相似度达99.37%。结合生理生化性状鉴定结果,可确定7株分离菌中3株为芽孢杆菌科芽孢杆菌属成员,4株为芽孢杆菌科赖氨酸芽孢杆菌属成员。The rumen fluid of sika deer was bathed at 85 ℃ for 20 min, then inoculated in the common nutrient agar to picked up the colony.After the colony purification and culture, the morphological characteristics of the individual and colony were observed. The physiological and biochemical characteristics of isolated strains were identified,and the phylogenetic tree was constructed by sequencing the bacterial 16 S rRNA. The results showed that the sequence similarity between 2 isolated strains and Bacillus subtilis was more than 99.93%, and the sequence similarity between one strain and B. licheniformis was 99.76%, the sequence similarity between 3 strains and Lysinibacillus macroides was 99.58%, and the sequence similarity between one strain and L. xylanilyticus was 99.37%.Combined with the physiological and biochemical identification results, it was confirmed that 3 out of 7 isolated strains belonged to Bacillus of Bacillaceae, and the remaining 4 strains belonged to Lysinibacillus of Bacillaceae.
