摘 要:收集63支鹿角,利用酚-氯仿法、改良酚-氯仿法、DNeasy~? Blood&Tissue Kit (50)、QIAamp~? DNA Investigator Kit (50)提取鹿角DNA,通过观察琼脂糖凝胶电泳结果,确定提取鹿角DNA的取样部位和提取方法。结果表明,鹿角中部DNA含量高于基部,更适合用于鹿角DNA提取工作;酚-氯仿法优于改良酚-氯仿法,DNeasy~? Blood&Tissue Kit (50)提取效果优于其他方法。进行鹿角DNA提取时,建议在鹿角中部取样,用DNeasy~? Blood&Tissue Kit (50)提取DNA。The methods〔Phenol-chloroform method, Modified phenol-chloroform method, DNeasy■ Blood & Tissue Kit(50) method,QIAamp■ DNA Investigator Kit(50) method〕were respectively used to extract DNA from 63 antlers. By observing the results of agarose gel electrophoresis, the sampling site and extraction method for antler DNA extraction were determined. The results showed that the DNA content in the middle of antler was higher than the base, which was more suitable for DNA extraction of antler. The phenol-chloroform method was superior to the modified phenol-chloroform method, and the DNeasy~? Blood & Tissue Kit(50) was better than other methods. For antler DNA extraction, it was recommended to take samples in the middle of the antler and extract DNA using the DNeasy~? Blood & Tissue Kit(50).
