摘　　要：目的 探讨血管紧张素Ⅱ对子宫内膜组织中的基质细胞(ESCs)数量和分泌功能的影响。方法 刮取人正常子宫内膜组织并分离其基质细胞。将细胞分为正常组(只加培养液)和实验组[培养液和不同浓度的血管紧张素Ⅱ(AngⅡ)],采用噻唑兰染色反映活细胞的数量;用酶联免疫法测出各组细胞悬液中Ⅰ型胶原和纤黏连蛋白的分泌情况。免疫印迹法比较各组细胞胞质中的金属基质蛋白酶9(MMP-9)和金属基质蛋白酶抑制剂-1(TIMP-1)的含量。结果 AngⅡ可以促进ESCs增殖,并呈剂量依赖性;使细胞分泌Ⅰ型蛋白(ColⅠ,0.741 2±0.052 1)和纤连蛋白(FN,0.723 6±0.032 8)的量增多,AngⅡ(10-5 mol/L)组细胞内MMP-9(0.397±0.047)含量明显减低,而TIMP-1(0.723±0.035)含量明显升高;与对照组比较,其差异有统计学意义(P<0.05)。结论 AngⅡ能使子宫内膜ESCs增殖,并增加其活性。[Objective]To explore the effect of angiotensin Ⅱ on the quantity and secretory functionhuman endometrial stromal cells(ESCs).[Methods]The ESCs was isolated from the human normal endometrial tissue. The cells were divided into normal group(culture medium only) and experimental group(culture medium and different concentration of angiotensin Ⅱ). Thiazolam staining was used to reflect the number of living cells,and enzyme-linked immunosorbent assay was used to detect the secretion of type I collagen and fibronectin in cell suspension of each group. The contents of matrix metalloproteinase-9(MMP-9) and tissue inhibitor of metalloproteinase-1( TIMP-1) in cytoplasm were compared by Western blot.[Results] Angiotensin Ⅱ could promote the proliferation of ESCs in a dose-dependent manner. The level of Col I(0.741 2±0.052 1)and fibronectin(FN)(0.723 6±0.032 8) while the content of MMP-9(0.397±0.047) decreased significantly and the content of TIMP-1(0.723±0.035) increased significantly in10-5 mol/L AngⅡ group,and the differences were statistically significant(P<0.05).[Conclusion]Angiotensin Ⅱcan proliferate endometrial stromal cells and increase their activity.
