摘  要:目的:探讨p38丝裂原活化蛋白激酶(p38 MAPK)在百草枯中毒急性肺损伤中对肿瘤坏死因子-α(TNF-α)的影响.方法:36只SD大鼠按随机数字法分为生理盐水组(NS组)、PQ中毒组(PQ组)、阻滞剂干预组(PQ+SB组),每组各12只,每组再从中各取第1天和第3天6只样本检测,通过观察肺组织病理学变化、肺泡灌洗液中炎症介质TNF-α变化,qRT-PCR法检测各组大鼠肺组织中TNF-α和p38 MAPK mRNA表达变化,以此来寻找环境压力应激反应蛋白p38 MAPK与肺组织炎症反应的相关性.结果:同一时间点的PQ组肺泡灌洗液中的TNF-α蛋白表达量显著高于NS组(P<0.05),而同一时间点PQ+SB组较PQ组肺泡灌洗液中的TNF-α蛋白表达量在第3天下降(P<0.05).同一时间点的PQ组TNF-α和p38 MAPK的mRNA表达量显著高于NS组(P<0.05),同一时间点PQ+SB组较PQ组表达量下降(P<0.05).而PQ组内不同时间点的肺组织中的TNF-α和p38 MAPK mRNA随时间增高(P<0.05);但PQ+SB组的TNF-αmRNA则无显著差异(P>0.05);肺组织病理学变化也显示同一时间点PQ组较NS组病理损伤明显,而PQ+SB组较PQ组病理损伤减少.结论:p38 MAPK抑制剂SB203580能够降低百草枯中毒大鼠急性肺损伤时的炎症介质TNF-α的表达,改善肺组织的炎症反应.Objective To investigate the effect of p38 mitogen-activated protein kinase (p38 MAPK) on tumor necrosis factor-α(TNF-a) in acute lung injury induced by paraquat poisoning.Methods Thirty-six SD rats were randomly divided into normal saline group (NS group), PQ poisoning group (PQ group) and blocker intervention group (PQ+SB group), with 12 rats in each group. Six samples were taken on day 1 and day 3. The pathological changes of lung tissue and the changes of inflammatory mediator TNF-αin alveolar lavage fluid were observed. qRT-PCR was used to detect TNF-α and p38 MAPK mRNA in lung tissue of each group. The expression change were used to find correlations between the environmental stress-stress protein p38 MAPK and lung tissue inflammation.Results The expression of TNF-α protein in the alveolar lavage fluid of the PQ group point was significantly higher than that of the NS group at the same time point (P < 0.05). The expression of TNF-α protein in the alveolar lavage fluid of the PQ+SB group compared with the PQ group at the third day and the amount of expression decreased (P < 0.05). The mRNA expression levels of TNF-α and p38 MAPK in PQ group were significantly higher than those in NS group at the same time point (P < 0.05). At the same time point, the expression of PQ+SB group was lower than that of PQ group (P < 0.05). The TNF-α and p38 MAPK mRNA in lung tissue at different time points in PQ internal group increased with time (P > 0.05), but there was no significant difference in the mRNA expression levels of TNF-αin PQ+SB internal group at the different time point(P > 0.05). Pathological changes of lung tissue also showed that the PQ group had more obvious pathological damage than the NS group at the same time point, while the PQ+SB group had less pathological damage than the PQ group at the same time pointconclusion p38 MAPK inhibitor SB203580 can reduce the expression of inflammatory mediator TNF-α and improve the inflammatory response of lung tissue in rats with acute lung injury induced by para
