红景天苷对子宫内膜上皮细胞的抗炎和保护作用

摘　　要：目的:探讨红景天苷对子宫内膜上皮细胞的抗炎和保护作用。方法:将细胞株分为空白对照组,正常对照组,红景天苷低(1×10^(-9) mol/L)、中(1×10^(-6) mol/L)及高(1×10^(-3) mol/L)剂量组。正常对照组,红景天苷低、中及高剂量组细胞均在内毒素(LPS)环境下培养48 h,采用噻唑蓝(MTT)法分析细胞增殖情况,采用酶联免疫吸附法测定白细胞介素6(IL-6)、白细胞介素1β(IL-1β)及肿瘤坏死因子α(TNF-α)水平。结果:24、48 h后,红景天苷中、高剂量组的细胞增殖水平明显高于空白对照组和正常对照组,差异均有统计学意义(P<0.05);24、48 h后,正常对照组,红景天苷低、中及高剂量组的IL-6、IL-1β及TNF-α水平明显高于空白对照组,差异均有统计学意义(P<0.05),且红景天苷剂量越高,IL-6、IL-1β及TNF-α水平越低。结论:红景天苷可保护炎症反应下的子宫内膜上皮细胞,显著减轻炎症反应。OBJECTIVE: To probe into the anti-inflammatory and protective effects of salidroside on endometrial epithelial cells. METHODS: The cell strains were divided into blank control group, normal control group, low-dose(1×10^-9 mol/L) group, medium-dose(1×10^-6 mol/L) group and(1×10^-3 mol/L) high-dose group of salidroside. Normal control group, low-dose group, medium-dose group and high-dose group were cultured in endotoxin(LPS) for 48 h, MTT method was adopted to analyze cell proliferation, interleukin-6(IL-6), interleukin-1β(IL-1β) and tumor necrosis factor α(TNF-α) levels were determined by enzyme-linked immunosorbent assay. RESULTS: After 24 h and 48 h, the cell proliferation level of medium-dose group and high-dose group were significantly higher than that of blank control group and normal control group, with statistically significant differences(P<0.05);after 24 h and 48 h, th IL-6, IL-1β and TNF-α levels of normal control group, low-dose group, medium-dose group and high-dose group were significantly higher than those of the blank control group, with statistically significant differences(P<0.05), and the higher the dose of salidroside, the lower the levels of IL-6, IL-1β and TNF-α. CONCLUSIONS: Salidroside can protect endometrial epithelial cells under inflammatory reaction, and significantly reduce inflammatory response.
