摘 要:目的:探讨miRNA-145和VEGF在儿童川崎病中的表达水平及其临床意义。方法:共纳入27例急性期川崎病的患儿,并依据心脏超声结果分为:伴有冠状动脉损伤组(CAL组)12例,无冠状动脉损伤组(NCAL组)15例。同期匹配18例健康儿童作为对照组。分别采用实时荧光定量PCR及ELISA法方法监测血浆中miRNA-145和VEGF在川崎病患儿CAL组、NCAL组及正常儿童血浆中的表达水平。结果: RT-PCR结果提示,三组miRNA-145的差异有统计学意义( F =64.53, P <0.01),CAL组患儿相对表达量低于NCAL组及正常对照组,NCAL组介于两组之间,差异具有统计学意义;ELLISA法提示三组间VEGF差异有统计学意义( F =52.87, P <0.01),其中CAL组患儿表达量明显高于NCAL组与正常对照组,NCAL组介于两组之间,差异有统计学意义。相关分析发现, miRNA-145和VEGF表达呈明显负相关关系( r =-0.707, P <0.01)。结论: miRNA-145可能通过调节VEGF的表达参与川崎病的发病机制,且与冠脉损伤的发生相关,miRNA-145和VEGF可作为川崎病辅助诊断指标。Objective:To investigate the expression and clinical significance of miRNA-145 and VEGF in children with Kawasaki disease(KD).Methods:A total of 27 children diagnosed as Kawasaki disease were divided into coronary artery injury(CAL) and without coronary artery injury(NCAL) based on the results ofechocardiograph. 18 healthy children were included as a control group.The plasma levels of miRNA-145 and VEGF in plasma of children with Kawasaki disease were examined by real-time fluorescence quantitative PCR and ELLIA method, respectively.Results:RT-PCR results showed that miRNA-145 was different between the three groups(F=64.53,P<0.01).The relative expression of miRNA-145 in CAL group was lower than that of NCAL group and the normal control group.NCAL group between the two groups.The difference was statistically significant. The ELLISA method suggested that VEGF also had a difference between the three groups(F=52.87,P<0.01).The expression of CAL group was significantly higher than that of NCAL group and the control group.No coronary artery injury group between the two groups. The difference was statistically significant.Correlation analysis showed that miRNA-145 and VEGF expression were significantly negative correlation(r=-0.707,P<0.01).Conclusion:miRNA-145 may be involved in the pathogenesis of Kawasaki disease by regulating the expression of VEGF, and it is associated with the occurrence of coronary artery injury. miRNA-145 and VEGF can be used as auxiliary diagnostic indicators for Kawasaki disease.
