摘　　要：目的探讨人类肝型脂肪酸结合蛋白(L-FABP)基因单核苷酸多态性(SNP)与胆囊结石病的关系。方法通过采集88例胆囊结石患者和154例健康者的静脉血,提取外周血基因组DNA组,检测L-FABP基因4个SNP位点。结果发现女性在rs1545224位点上,疾病组中A∶G的值高于对照组(P<0.01)。SNP位点rs1545224、rs2919867和rs2197076的基因型频率与胆囊结石疾病具有相关性(P<0.05)。在共显性模型中,rs1545224位点AA和GG基因型的女性人群患有胆囊结石的危险度分别是AG基因型人群的2.93和2.31倍;在超显性模型中,该位点AA/GG基因型患胆囊结石的危险度是AG基因型2.542倍。同样的结果也在rs2919867(P<0.01)和rs2197076(P<0.01)位点中发现。多因子降维模型分析交互作用提示L-FABP基因的rs1545224位点与早餐情况与体育锻炼这2个环境因素有关。结论L-FABP基因中rs1545224、rs2919867和rs2197076的多态性位点可能与女性患者胆囊结石的易感性有关。Objective To investigate the relationship among single nucleotide polymorphism ( SNP), human liver fatty acid binding protein ( L - FABP) and gallstone. Methods The genomic DNA of peripheral blood was extracted from 88 patients with gallstone and 154 healthy people, and the genotype of L - FABP polymorphism was analyzed by double deoxidization chain termination method. Results We found that in women the value of A∶G in the case group was higher than that in the control group at the rs1545224 site ( P < 0. 01). SNP rs1545224, rs2919867 and rs2197076 were correlated with gallstones at SNP site ( P < 0. 05). At the co - dominant model, risky of AA and GG genotypes were 2. 93 and 2. 31 times greater than AG genotype in women suffering from gallstones. In addition, the over - dominance model, risky of AA /GG genotypes was 2. 542 times greater. The rs2919867 ( P < 0. 01) and rs2197076 ( P < 0. 01) were also found;Multifactor dimensionality reduction model analysis found rs1545224 locus of L - FABP gene was related to the two environmental factors of breakfast and physical exercise. Conclusion The polymorphisms of rs1545224, rs2919867 and rs2197076 on L - FABP may be related to female patients with gallstone.
