摘 要:在单因素试验的基础上,运用响应面分析方法优化酸性染料法测定走马胎生物碱的最佳条件,并建立数学模型验证其可靠性.回归方程极显著(p=0.0023),而失拟项不显著(p=0.3794),预测值与实际值吻合(R2=0.9717);影响测定走马胎生物碱因素的重要性大小顺序为:缓冲液pH值>静置时间>酸性染料用量;酸性染料法测定走马胎生物碱含量的最佳条件为:静置时间2 h,酸性用量7 ml,缓冲液pH=7.20;在此优化条件下,以同批次的走马胎为测定对象,验证该方法的精密度、重复性、准确度,其RSD值为3.66%,平均加标回收率为97.99%,加标回收率的RSD值为4.23%.因此,该法简便、快速、准确度高且重现性、精密度较好,可以作为测定走马胎生物碱的方法.Based on single factor tests, the response surface method was used to optimize the determination condition of total alkaloids in Ardisia gigantifolia Stapf by acid dye colorimetry. The quadratic term model of the determination conditions was established and the reliability was checked. The results of regression analysis presented a extremely significant regression equation( p =0.0023), a non -significant lack of it(p=0.3794)and a high coefficient of determination value(R^2) of 0. 9717 for the response surface model,which showed a satisfactory agreement of the polynomial regression equation with experimental data. The influence of three factors followed the sequence as: buffer solution pH>standing time>acid dye colorimetry amount. The optimum determination conditions of Ardisia gigantifolia Stapf’s total alkaloids using acid dye colorimetry were standing time: 2 h, acid dye colorimetry amount: 7 mL, buffer solution pH: 7.20. According to the optimum determination conditions, the precision, repeatability and accuracy of the method were verified using Ardisia gigantifolia Stapf. The relative standard deviations of total alkaloids contents in the Ardisia gigantifolia Stapf and recovery rate is 3.66% and 4.23%, respectively, and the average recovery rate was 97.99%. The study provided a reliable method for accurate, efficient and cost effective detection of total alkaloids contents in Ardisia gigantifolia Stapf.
