摘 要:目的观察IFN-γ预处理同种异体骨髓间充质干细胞(BMSCs)复合脱钙骨基质(DBM)修复新西兰兔桡骨骨缺损的能力。方法将48只新西兰大白兔随机分为四组,各12只,均制作单侧桡骨缺损模型。BMSCs体外培养扩增,用IFN-γ预处理,成骨诱导。采用BMSCs和DBM构建组织工程骨修复兔桡骨缺损。观察术后各时间点骨修复情况,并检测BMSCs的ALP活性。结果术后12周,A组出现少量骨痂,但未形成完整连续骨痂;B组部分松质骨被吸收,骨折断面处有少量新生骨,骨痂形成不佳,骨缺损不能修复,缺损区由纤维组织填充;C组新生骨量少于D组,且没有达到全部骨性愈合;D组在骨缺损区域出现比较完整成熟的骨组织结构,骨小梁排列趋于有序,在骨与材料交界断端形成连续的骨桥。术后12周,C、D组抗弯曲能力明显高于A、B组,且D组高于C组(P<0.05)。成骨诱导组及IFN-γ预处理组的ALP活性均高于BMSCs组(P<0.05)。结论IFN-γ预处理同种异体BMSCs联合DBM修复兔桡骨缺损能显著促进骨形成,改善桡骨缺损修复效果。Objective To observe the ability of allogeneic bone marrow stromal cells(BMSCs) with IFN-γ pretreatment combined with decalcified bone matrix(DBM) on repair of radius defect in rabbits. Methods A total of 48 New Zealand white rabbits were randomly divided into 4 groups, with 12 in each group. Unilateral radial defect models were made in all rabbits. BMSCs were cultured in vitro and pretreated with IFN-γ for osteogenic induction. BMSCs and DBM were used to construct tissue engineered bone for repairing radial defects in rabbits. The restoration were observed at various time points after operation, and the ALP activity of BMSCs were examined. Results At 12 weeks after operation, a small amount of callus appeared in the group A, but no complete and continuous callus was formed;some cancellous bone was absorbed in the group B, and a small amount of new bone was formed at the fracture section, the callus formation was not good, the bone defect could not be repaired, and the defect area was filled with fibrous tissue;the new bone in the group C was less than that in the group D, and did not achieve all bone healing;in the group D, the mature bone structure appeared in the bone defect area, the trabecular arrangement tended to be orderly, and the continuous bone bridge formed at the bone-material junction. The bending resistance of the groups C and D were significantly higher than those of the groups A and B, and that of the group D was significantly higher than the group C(P <0.05). The ALP activity in the osteogenic induction group and IFN-γ pretreatment groups were higher than those in the BMSCs group(P <0.05). Conclusion The allogeneic BMSCs with IFN-γ pretreatment combined with DBM on repair of radius defect in rabbits can significantly promote the bone formation, and improve the repair of radius defects.
