改良微柱凝胶法的建立及其在检测嵌合体ABO血型中的应用

摘　　要：目的建立改良微柱凝胶法用于嵌合体ABO血型的检测。方法建立改良微柱凝胶法,用于检测A(或B)和O型红细胞不同比例混合红细胞与盐水试管法检测为混合外观或微柱凝胶法检测为双群现象的标本,用散在红细胞抗原验证试验验证。结果改良微柱凝胶法混合红细胞检测限为20∶1至1∶21,10例标本2例为双群,散在红细胞抗原验证试验标本中存在2种红细胞,8例为非双群,散在红细胞抗原验证试验为均一红细胞;微柱凝胶法混合红细胞检测限为10∶1至1∶15,10例标本均为双群;盐水试管法检测10例标本6例为混合外观。结论改良微柱凝胶法提高了混合红细胞中微量细胞的检出能很好地将嵌合体血型与抗原减弱型和ABO亚型区分开来,是一种适用于检测嵌合体血型的有效方法。Objective A modified microcolumn gel method for determination of ABO blood group in chimera was established. Methods A modified microcolumn gel method was established for the detection of 1% A( or B),O type mixed red blood cells and saltwater test tube method for specimens with mixed appearance or microcolumn gel method for the detection of double-cluster phenomena,which was verified by the test of scattered erythrocyte antigen. Results The modified microcolumn gel method for mixed red blood cell detection was limited to 20∶1-1∶21. In 11 cases,3 cases were double groups and the scattered erythrocyte antigen validation test showed two types of erythrocytes. 8 specimens were not two-group and the scattered erythrocyte antigen validation test showed single type of erythrocyte. Microcolumn gel method mixed red blood cell detection limit was 10∶1-1∶15. The 11 specimens were all double groups. 10 cases of 6 specimens were mixed appearance by salt tube test. Conclusion The improved microcolumn gel method improves the detection of trace cells in mixed red blood cells, and can distinguish the chimera blood type from the antigen weakening type and ABO subtype. It is an effective method for detecting chimera blood type.
