微压香氧预处理对MCAO大鼠Apaf-1表达的影响

摘　　要：目的探讨微压香氧(Micro-Hyperbaric Oxygen Aromatherapy,MHBO-AT)预处理对MCAO大鼠脑梨形皮质Apaf-1表达的影响。方法将雄性成年SD大鼠85只随机分为假手术组15只、模型组35只、微压香氧预处理组35只。除假手术组外其余各组根据再灌注后时间点不同又分为6h、12h、24h3个亚组,6h、12h每组10只,24h组15只。观察24h脑组织TTC染色,各时间点免疫组织化学法和Western blot检测大鼠脑梨形皮质凋亡酶激活因子(Apaf-1)的表达。结果24h TTC染色显示与模型组比较,微压香氧预处理组梗死体积明显缩小,差异有显著性(P<0.05)。免疫组织化学法和Western blot显示与模型组相比,微压香氧预处理组各时点Apaf-1蛋白表达均降低(P<0.05)。结论MHBO-AT预处理对大鼠脑缺血再灌注损伤产生神经保护作用,通过抑制脑梨形皮质中Apaf-1蛋白的表达可能是其作用机制。Objective To investigate the effect of Micro-Hyperbaric Oxygen Aromatherapy on the expression of Apaf-1 in the brain piriform cortex of MCAO rats.Methods 85 male SD rats were randomly divided into 15 sham operation group,35 model group and 35 model + micro-hyperbaric oxygen aromatherapy group.The rats in model group and model + micro-hyperbaric oxygen aromatherapy group were further divided into 3 subsets,according to after reperfusion time of 6 h,12 h,and 24 h,10 in each subset at 6 h and 12 h,and 15 in 24 h subset.The TTC staining of brain tissue was observed at 24 h,and the expression of Apaf-1 in rat piriform cortex was detected by Immunohistochemistry and Western blot at each time point.Results 24 h TTC staining showed that compared with the model group,the infarct volume of the micro-hyperbaric oxygen aromatherapy group was significantly reduced,and the difference was significant(P<0.05).Immunohistochemistry and Western blot showed that compared with the model group,the number of Apaf-1 protein in the micro-hyperbaric oxygen aromatherapy group decreased at each time point(P<0.05).Conclusion MHBO-AT has neuroprotective effects on cerebral ischemia-reperfusion injury in rats.It may be the mechanism by inhibiting the expression of Apaf-1 protein in the piriform cortex.
