摘  要:目的:研究并探讨月季花总黄酮对小鼠反复脑缺血再灌注模型NO、NOS水平和脑水肿的影响。方法:制造反复脑缺血再灌注模型小鼠,将小鼠分为假手术组、模型组、脑络通组(0.75 g/kg)和月季花总黄酮高、中、低剂量组(0.4 g/kg、0.2 g/kg、0.1 g/kg),共6组,连续给药7 d,于第7 d灌胃1 h后,麻醉小鼠并仰卧固定,分离双侧颈总动脉(CCA)用微动脉夹夹闭10 min,松开10 min,再夹闭10 min复制小鼠反复脑缺血再灌注模型,给予适量青霉素后缝合伤口,于第8 d早上给药1 h后观察各组动物死亡情况,进行摘眼球取血,低温离心测定血清中NSE水平,断头冰盘剥脑,矢状切取一半匀浆制成10%脑组织匀浆液,检测NO、NOS水平,另一半称重后放置于烘箱中105℃烘至恒重,计算脑组织含水量。结果:小鼠反复脑缺血再灌注模型复制成功。与模型组相比,月季花总黄酮各组小鼠死亡率不同程度降低;月季花总黄酮高剂量组小鼠血清中NSE含量显著降低(P<0.01),中剂量和低剂量组可明显降低小鼠血清中NSE含量(P<0.05);月季花总黄酮高剂量可显著降低脑组织匀浆中NO、NOS水平(P<0.01),中剂量组可明显降低脑组织匀浆中NO、NOS水平(P<0.05),低剂量组可明显降低NOS活性(P<0.05);月季花总黄酮各组对脑组织含水量有明显改善作用,其中高剂量组和中剂量组可降低小鼠脑组织含水量(P<0.05)。结论:月季花总黄酮可显著降低小鼠反复脑缺血再灌注后血清中NSE含量和脑组织NO含量,抑制NOS活性,明显改善小鼠缺血后脑水肿,减轻小鼠反复缺血再灌注损伤。Objective: To investigate the effect of the total flavonoids of China rose on NO, NOS and hydrocephalus in repeated CIR mice. Methods: The model of cerebral ischemia was established by repeated cerebral ischemia and reperfusion(CIR) in mice. The mice were randomly divided into the sham operation group, the model group, the Naoluotong group(0.75 g/kg), and the total flavonoids groups of low-dose, medium-dose and high-dose(0.1 g/kg, 0.2 g/kg, 0.4 g/kg). The medication was continuous 7 days;bilateral common carotid arteries(CCA) were separated and clipped 10 min followed by 10 min unclipped and 10 min clipped to set up repeated CIR model 1 h after administration on 7th day. After suturing the wound treated by penicillin, 1 hour after administration on the morning of the eighth day, death of the animal was observed. Blood sample and brain tissue sample were collected to detect serum level of NSE and expressions of NO and NOS. The water contents were also measured in brain tissue. Results: The model of repeated CIR was made successfully in mice. Compared to those in the model group, the mortality rates were decreased in the total flavonoids groups;the serum content of NSE was significantly lowered in the total flavonoids group of high- dose( P <0.01);it was also lowered in the medium-dose group and low-dose group( P <0.05);the expressions of NO and NOS were significantly decreased in the high-dose group( P <0.01), the decrease was also seen in the medium-dose group( P <0.05), and the activity of NOS was reduced in the low-dose group (P <0.05);the total flavonoids had significant effects on water contents of brain tissue, and the water contents were greatly reduced in the high-dose group and in the medium-dose group( P <0.05). Conclusion: The total flavonoids of China rose can significantly reduce serum content of NSE and brain content of NO and inhibit the activity of NOS in the repeated CIR mice, it can improve the hydrocephalus after ischemia and reduce the injury of repeated CIR.
