摘  要:目的探讨miR-200a通过调控低氧诱导因子-1α(HIF-1α)与血管内皮生长因子(VEGF)通路调节鼻息肉上皮细胞的增殖及凋亡作用。方法鼻息肉上皮CNE细胞分别转染miR-200a模拟物及对照模拟物,采用qRT-PCR法检测miR-200a表达水平,采用MTT法与Tunel试验测定细胞增殖及凋亡作用,采用Western blot法测定HIF-1α与VEGF表达水平。结果 qRT-PCR检测显示经过转染,在miR-200a表达水平上,实验组较对照组、空白组升高约37. 82倍。转染48 h后,实验组的活细胞显著低于对照组与空白组;细胞凋亡率方面,实验组和对照组以及空白组对比差异均有统计学意义(P <0. 05);转染48 h后,经过Western blot检测得出,在HIF-1α和VEGF表达水平方面,实验组比对照组、空白组明显降低(P <0. 05)。结论 miR-200a可通过激活HIF-1α与VEGF通路,限制鼻息肉上皮细胞的增殖能力,提升凋亡能力,进而影响鼻息肉细胞生物行为。Objective To investigate the effects of miR-200a regulates the proliferation and apoptosis of nasal epithelial cells by regulating hypoxia inducible factor-1α(HIF-1α) and vascular endothelial growth factor (VEGF) pathway. Methods Nasal polyps epithelial CNE cells were transfected miR-200a mimic and control,qRT-PCR method was used to detect the expression level of miR-200a,MTT method and Tunel test were used to detect the cell proliferation and apoptosis,the level of HIF-1ɑ and VEGF expression were determined by Western blot. Results The results of qRT-PCR showed that the expression level of miR-200a in the experimental group were higher than that in the blank group and the negative control group. After transfection of 48 h,the proportion of live cells in the experimental group were lower than the negative control group and blank group,there was statistically significant difference compared among the groups ( P <0.05). Western blot assay showed that transfection after 48 h,the expression level of HIF-1ɑ and VEGF in the experimental group were significantly lower than the negative control group and blank control group ( P <0.05). Conclusion miR-200a can inhibit the proliferation of nasal polyps epithelial cells by increasing HIF-1 and VEGF pathway,and improve the ability of apoptosis.
