摘 要:目的观察复方苦参注射液对放射后鼻咽上皮细胞(NP69)存活的影响,并探讨其放射防护作用机制:方法采用四甲基偶氮唑盐(MTT)法检测电离辐射后NP69细胞的增殖情况;采用流式细胞法观察复方苦参注射液对电离辐射后NP69细胞凋亡的影响;克隆形成试验检测各组细胞的克隆形成率;采用DCFH-DA荧光探针检测复方苦参注射液对电离辐射后NP69细胞氧自由基的清除作用。结果苦参干预组NP69细胞的增殖率高于单纯放射组(P<0.05);苦参干预组NP69细胞的凋亡率较单纯放射组明显减少(P<0.05);苦参干预组NP69细胞克隆形成率高于单纯放射组(P<0.05);经复方苦参注射液处理后,NP69细胞氧自由基活性较单纯放射组明显降低(P<0.05)。结论复方苦参注射液可能是通过清除NP69细胞中辐射产生的氧自由基,从而抑制辐射后NP69细胞凋亡并促进其增殖。Objective To investigate the effect of compound Kushen injection on the survival of nasopharyngeal epithelial cells(NP69) after radiation, thus to explore its radioprotective mechanism. Methods Methylthiazolyl tetrazolium(MTT)assay was used to observe effect of compound Kushen injection on the proliferation of NP69 cells after radiation. Flow cytometry was used to detect apoptosis rate of NP69 cells. The clone formation rates of cells in various groups were detected by clone formation assay. DCFH-DA fluorescence probe was used to detect the level of oxygen radicals. Results After radiation,compared with the radiation alone group,the proliferation rate of NP69 cells in Kushen intervention group was higher(P<0.05), with lower apoptosis rate(P<0.05), higher clone formation rate(P<0.05), and decreased level of oxygen radicals(P<0.05). Conclusion Compound Kushen injection has effects on inhibiting the apoptosis and promoting the proliferation of NP69 cells after radiation by clearing oxygen radicals induced by radiation.
