摘 要:利用现代分子标记技术探索了影响猪产仔数的候选基因CATSPER1多态性,并分析了多态性与产仔数间的相关性,为建立产仔数分子标记辅助选择方法提供依据。试验采用PCR-RFLP(HhaI限制性内切酶)技术,分析了264头定远猪母猪的CATSPER1基因exon 1的c.A779G多态性,并采用最小二乘法分析了CATSPER1基因多态性对产仔数影响的遗传效应。结果显示,CATSPER1基因c.A779G位点在定远猪中存在多态性,AA、AG和GG型频率分别为0.030 3、0.272 7和0.697 0,野生型等位基因A的基因频率为0.166 7,经检验该位点在定远猪群中处于哈德-温伯格平衡状态(P>0.05);AA型个体的总产仔数和产活仔数最高,GG型个体最低,AA型总产仔数均极显著高于AG和GG型(P<0.01),AA型个体的产活仔数均显著高于AG和GG型(P<0.05),而AG和GG型个体间的总产仔数和产活仔数均差异不显著(P>0.05);遗传效应分析表明,等位基因A为定远猪优良等位基因,A等位基因替代G等位基因的效应均为正值,因此选择AA型个体留种有利于提高群体的产仔数。The polymorphism of the candidate gene CATSPER1 of pig litter size was explored by modern molecular marker technology.The correlation between polymorphism and litter size was analyzed,which provide a basis for establishing molecular marker-assisted selection method for litter size.In this study,PCR-RFLP(HhaI restriction enzyme)technology was used to analyze the c.A779G locus polymorphism of CATSPER1 gene in 264 Dingyuan sows,and the genetic effects of CATSPER1 gene polymorphism on litter size were analyzed by least square method.The results showed that the c.A779G locus of CATSPER1 gene was polymorphic in Dingyuan pigs.The frequencies of AA,AG and GG were 0.030 3,0.272 7 and 0.697 0 respectively.The frequency of wild-type allele A was 0.166 7.The locus was in the Had-Weinberg equilibrium in Dingyuan pig herd(P>0.05).The total number born(TNB)and the number born alive(NBA)of AA-type individuals were the highest respectively,GG-type individuals were the lowest.AA-type individuals were extremely significantly higher than AG and GG-type individuals with the total litter size(P<0.01)and significantly higher than AG and GG-type individuals with the live litter size(P<0.05).However,there was no significant difference in total litter size and live litter size between AG and GG-type individuals(P>0.05).Genetic effects analysis showed that allele A was a good allele in Dingyuan pigs,and the genetic effect of allele A replacing allele G was positive.Therefore,we should select more AA type individuals for boars or sows to raising litter size.
