摘  要:目的:研究牡丹皮炭中多成分的整合药代动力学与止血药效作用的相关性,阐明牡丹皮炭的止血作用机制及其炮制机理。方法:大鼠分为空白组、模型组和给药组,模型组采用血热出血模型,各组于给药后不同时间点分别采用HPLC方法检测多个成分的血药浓度;采用酶联免疫吸附法(ELISA)法测定TXB_2和6-Keto-PGF_(lα)值;结合AUC自定义权重系数,计算牡丹皮炭多成分整合药代动力学浓度,并与止血药效指标相结合,进行量-效-时相关性分析。结果:牡丹皮炭多成分整合血药浓度随时间变化与TXB_2和TXB_2/6-Keto-PGF_(lα)值呈正相关。结论:牡丹皮炭多成分整合药代动力学与药效作用相关研究具有一定的科学性和可行性,可用于牡丹皮炭止血作用机制及其炮制机理的阐述。Objective:To study the correlation between the multicomponent pharmacokinetics and pharmacodynamic effects of Moutan Cortex charcoal,and to elucidate the hemostatic mechanism and processing mechanism.Methods:Rats were divided into blank group,model group and drug-administered group.The model group adopted the blood hot hemorrhage model.The plasma concentrations of multicomponent components were detected by HPLC at different time.The enzyme-linked immunosorbent assay(ELISA)method was used to determine the TXB2 and 6-Keto-PGFlα values.The integrated concentrations were calculated according to the AUC self-defined weighting coefficiency,then the potential relationship between the integrated concentration and the hemostatic efficacy was analyzed.Results:The multicomponent integrated plasma concentration of Moutan Cortex charcoal was positively correlated with TXB2 and TXB2/6-Keto-PGFlα.Conclusion:The research on the relationship between multicomponent integrated pharmacokinetics and pharmacodynamic effects of Moutan Cortex charcoal is scientific and feasible,and also can elucidate the hemostatic mechanism and processing mechanism of Moutan Cortex charcoal.
