摘 要:目的:建立白及提取物HPLC指纹图谱,以LPS诱导RAW264.7巨噬细胞为炎症模型,考察各提取物对IL-1β、IL-6、TNF-α等炎症因子抑制活性,分析其谱效关系。方法:采用聚酰胺柱色谱和系统溶剂萃取等方法制备系列白及提取物,并建立其HPLC指纹图谱。以LPS诱导的RAW264.7巨噬细胞为炎症模型,采用CBA法及流式细胞术测定各提取物对炎症因子IL-1β、IL-6、TNF-α的抑制作用;通过灰色关联度分析法研究其谱效关系。结果:白及各提取物F1~F8,除了F5抗炎活性较差外,其余各部位均具有一定抗炎活性,其中F7和F3具有显著抗炎活性。白及醇提物18个特征峰中,对IL-1β、IL-6、TNF-α抑制活性关联度大于0.8的峰分别有7、7、4个,其中9号峰对各炎症因子抑制活性均表现出较强活性;通过制备、鉴定该峰对应化合物为贝母兰宁,并进一步确证了其抗炎活性。结论:本研究构建了白及抗炎谱效关系,并鉴定了其抗炎活性较强的药效成分,确证贝母兰宁为白及发挥抗炎活性的主要药效成分之一,这为进一步科学合理开发利用白及奠定了良好基础。Objective:To establish the HPLC fingerprint of the extracts from Bletilla striata,and to investigate its inhibitory activities on IL-1β,IL-6 and TNF-α using RAW264.7 macrophage inflammatory model induced by LPS.Methods:A series of Bletilla striata extracts were prepared by polyamide column chromatography and systematic solvent extraction method,and their HPLC fingerprints were established.RAW264.7 macrophages induced by LPS were used as inflammatory model.The inhibitory effects of each extract on inflammatory factors of IL-1β,IL-6 and TNF-α were determined by CBA and flow cytometric methods.The spectrum-effect relationship was studied by grey relational analysis.Results:Except for the poor anti-inflammatory activity of F5,all F1~F8 extracts of Bletilla striata had certain anti-inflammatory activities,F7 and F3 had significant inhibitory activities.Eighteen characteristic peaks in the Bletilla striata extracts were selected,and spectrum-effect relationship analysis indicated that there were seven,seven and four peaks with a grey relational grade greater than 0.8 of IL-1β,IL-6 and TNF-α inhibitory activities,respectively.Peak nine showed strong activity against all inflammatory factors,which was purified and identified as compound coelonin,and its anti-inflammtion activity was confirmed.Conclusion:The anti-inflammatory activity spectrum-effect relationship of Bletilla striata is established and its anti-inflammatory active ingredient is identified.It confirms that coelonin is one of the main effective component of Bletilla striata to play anti-inflammatory activity.This has laid a good foundation for the further scientific and rational exploitation and utilization of Bletilla striata.
