摘  要:目的:获取有效区分和鉴定黄草乌和展毛黄草乌的信息位点。方法:通过高通量测序技术以及相关数据分析软件对黄草乌和展毛黄草乌的叶绿体全基因组结构特征进行比较分析。结果:黄草乌和展毛黄草乌的叶绿体全基因组序列为长度155 761 bp和155 816 bp的环状四分体结构,均包含132个基因,包括85个蛋白编码基因(PCGs),37个转运RNA(tRNA)以及8个核糖体RNA(rRNA)。在黄草乌和展毛黄草乌的叶绿体全基因组的比较分析中:(1)分别鉴定出30条和47条重复序列,其中黄草乌在trnR-atpA基因间隔区上缺失1个34 bp的回文重复,展毛黄草乌在atpF-atpH基因间隔区上缺失1个38 bp的正向重复;(2)分别鉴定出282和280个SSRs,黄草乌的9个A碱基的单核苷酸重复位于trnL-ccsA基因间隔上,而展毛黄草乌的8个A碱基的单核苷酸重复位于ccsA基因上。通过最大似然法(ML)和贝叶斯推论(BI)构建系统发育树发现,2个变种与乌头亚属聚在一起,并以100%的支持率形成一个独立分支。结论:本研究结果丰富了乌头属物种的叶绿体全基因组资料,同时将对黄草乌种下变种及乌头属物种的分子标记开发和鉴定提供有效的科学数据。Objective:To obtain informative sites for intraspecific identification of Aconitum vilmorinianum.Methods:High-throughput sequencing technology and data analysis softwares were used to analyze the structures of chloroplast genome of Aconitum vilmorinianum and Aconitum vilmorinianum var.patentipilum.Results:The lengths of Aconitum vilmorinianum and Aconitum vilmorinianum var.patentipilum were 155 761 bp and 155 816 bp,respectively,with a typical quadripartite structure,a total of 132 genes were identified,including 85 protein-coding genes(PCGs),37 RNA(tRNA)genes and eight ribosomal RNA genes(rRNA).In addition,there were 30 and 47 repeat sequences in Aconitum vilmorinianum and Aconitum vilmorinianum var.patentipilum,respectively.A forward repeat with 38 bp long in the atpF-atpH intergenic was lost in Aconitum vilmorinianum var.patentipilum,and apalindromic repeat with 34 bp long in the trnR-atpA intergenic was lost in Aconitum vilmorinianum.Moreover,282 and 280 SSRs were identified in Aconitum vilmorinianum and Aconitum vilmorinianum var.patentipilum,respectively.Among these SSRs,a polyA with 9 bp long in the trnL-ccsA intergenic was discovered in Aconitum vilmorinianum,and a polyA with 8 bp long in the ccsA gene was found in Aconitum vilmorinianum var.patentipilum.Maximum Likelihood and Bayesian Inference cluster analysis showed that the two varieties were clustered in Aconitum Subgenus Aconitum,and formed an independent clade with 100% bootstrap support.Conclusion:The results will deepen our understanding on the chloroplast genomes of Aconitum species,and also provide effective molecular markers for the species identification of Aconitum vilmorinianum,its variety and Aconitum genus.
