摘 要:目的:建立一种快速预测远志药材中化合物tenuifoliside C含量高低的方法,用于远志药材镇静安神药效强弱的初步评价。方法:采用UPLC法测定远志药材中化合物tenuifoliside C的含量,并对远志药材进行ISSR分析,通过相关分析筛选出与化合物tenuifoliside C含量高低相关的ISSR特异性条带,针对该ISSR条带设计特异性PCR引物。结果:筛选出4条与化合物tenuifoliside C含量相关的ISSR特异性条带,对其中1条特异条带(UBC835-1)进行胶回收、克隆测序分析,设计出1对特异性PCR引物(YZ-TC),可用于快速鉴别tenuifoliside C含量较低的远志药材样品。结论:采用ISSR分子标记结合相关分析技术,所设计出的特异性PCR引物可用于远志药材样品中tenuifoliside C含量高低的快速预测。Objective:To establish a method for rapidly predicting the content of tenuifoliside C in Polygala tenuifolia in order to preliminaryly evaluate the sedative effects of Polygala tenuifolia.Methods:UPLC method was used to determine the content of tenuifoliside C in Polygala tenuifolia.ISSR analysis was carried out in Polygala tenuifolia.ISSR-specific bands related to the content of tenuifoliside C were screened out by correlation analysis,and the specific PCR primers were designed for this ISSR band.Results:Four ISSR specific bands related to the content of tenuifoliside C were screened out and one of the specific bands(UBC835-1)was gel-recovered and cloned and then sequenced.One pair of specific PCR primers(YZ-TC)was designed,which could be used to identify Polygala tenuifolia samples with lower tenuifoliside C content quickly.Conclusion:The PCR primers designed by ISSR molecular marker combined with correlation analysis technology can be used to predict the content of tenuifoliside C in Polygala tenuifolia rapidly.
