消风宣窍汤治疗小鼠变应性鼻炎血清代谢组学研究

摘　　要：目的:分析小鼠变应性鼻炎(AR)血清代谢谱变化,以及消风宣窍汤对AR的治疗效应和可能机制。方法:将44只BALB/c小鼠随机分为正常对照组、模型组、消风宣窍汤组、孟鲁斯特组,采用OVA致敏建立小鼠AR模型,利用GC-MS分析消风宣窍汤干预后对AR血清样本代谢谱的影响。结果:造模后,AR血清代谢谱发生了广泛而显著的变化,主要涉及苯丙氨酸等多种氨基酸的生物合成与代谢、花生四烯酸代谢等;消风宣窍汤干预后能逆转L-赖氨酸、L-组氨酸、花生四烯酸等16种代谢物的变化(P<0.05),且使AR血清代谢谱向正常对照组回归,而西药孟鲁斯特组无明显回归趋势。结论:AR是系统性疾病,消风宣窍汤可通过调控多种炎症、免疫相关的血清代谢物,使其趋于回归,从而发挥治疗AR的作用。Objective:To analyze the changes of serum metabolic profiles of allergic rhinitis(AR)in mice and the therapeutic effects and possible mechanisms of Xiaofeng xuanqiao decoction(XFXQ)on AR.Methods:Forty-four BALB/c mice were randomly divided into normal group,model group,XFXQ group and montelukast group.AR models were established by sensitization with OVA,and GC-MS was used to analyze the effect of XFXQ on the secrum metabolic profiles of serum samples of AR.Results:There were extensive and significant changes in the serum metabolic profiles of AR after building model,which involved the biosynthesis and metabolism of various amino acids such as phenylalanine and arachidonic acid metabolism.XFXQ could reverse the changes of sixteen metabolites such as L-lysine,L-histidine and arachidonic acid(P<0.05),and make the serum metabolic profile of AR returned to the normal nearly,while the montelukast group had no obvious regression trend.Conclusion:AR is a holistic disease,XFXQ can be used to treat AR by regulating various inflammatory and immune-related serum metabolites to make it return to normal.
