摘 要:目的:研究不同剂量资生汤加减对慢性萎缩性胃炎型(CAG)Wistar大鼠胃分泌功能(胃蛋白酶活性、胃液pH、胃液总酸度和胃结合黏液),血清一氧化氮合酶(NOS)、生长抑素(SS)、前列腺素E_2(PGE_2)、胃泌素(GAS)及水通道蛋白3(AQP3)、水通道蛋白4(AQP4)表达的影响,探讨资生汤治疗CAG的作用机制。方法:按随机数字表法从90只大鼠中取15只设正常对照组,不予造模;其余75只采用综合因素诱发建立CAG模型后分为模型组、维酶素组及资生汤高、中、低剂量组,均连续治疗60 d。检测胃分泌功能及血清NOS、SS、PGE_2、GAS水平和胃窦组织AQP3、AQP4蛋白表达。结果:与模型组比较,资生汤高、中剂量组胃蛋白酶活性、胃液总酸度、胃结合黏液、血清PGE_2、GAS水平及胃窦组织AQP3、AQP4蛋白表达显著升高(P<0.05或P<0.01),胃液pH及血清NOS、SS显著降低(P<0.05或P<0.01)。结论:高、中剂量资生汤能显著改善CAG大鼠的胃分泌功能及血清NOS、SS、PGE_2、GAS水平和胃窦组织AQP3、AQP4蛋白的表达。Objective:To observe the effect and mechanism of the action of Zishengtang modified in different doses on gastric secretory function(pepsin activity,pH of gastric juice,total acidity of gastric juice and gastric mucus)and the expressions of serum nitric oxide synthase(NOS),somatostatin(SS),prostaglandin E2(PGE2),gastrin(GAS),Aquaporin 3(AQP3)and Aquaporin 4(AQP4)in chronic atrophic gastritis(CAG)rats.Methods:15 rats were randomly selected into blank control group.The CAG model was induced by comprehensive factors in the other 75 rats,and the rats were randomly divided into model group,vatacoenayme group,Zishengtang low,middle-and high-dose groups.All rats were treated for 60 d.The function of gastric secretion and the protein expressions of NOS,SS,PGE2,GAS in serum,AQP3 and AQP4 in gastric antrum tissue were detected.Results:Compare with model group,the activity of pepsin,total acidity of gastric juice,acidity,PGE2 and GAS in serum,AQP3 and AQP4 in gastric antrum tissue of Zishengtang high and middle dose groups were increased significantly(P<0.05 or P<0.01).The pH in gastric juice,NOS and SS in serum were significantly decreased(P<0.05 or P<0.01).Conclusion:Zishengtang with middle and high dose can obviously improve the gastric secretion function and the expressions of NOS,SS,PGE2,GAS in serum,AQP3 and AQP4 in gastric antrum tissue of CAG rats.
