摘 要:目的:观察灯盏乙素对β-淀粉样蛋白(Aβ)诱导的神经母细胞瘤(SH-SY5Y)细胞中组蛋白H2A、H2B、H3表达的影响,探讨其针对阿尔茨海默病(Alzheimer′s disease,AD)发挥表观遗传方面治疗作用的可能性。方法:将SH-SY5Y细胞分为对照组、灯盏乙素处理组、Aβ处理组和Aβ+灯盏乙素不同剂量处理组,用CCK-8法检测细胞存活率;用流式细胞术测定各组细胞凋亡率;用蛋白质印迹法和实时定量聚合酶链法检测各组细胞H2A、H2B、H3蛋白和mRNA的表达。结果:与对照组比较,Aβ处理组细胞存活率显著降低,凋亡率显著升高,H2A、H2B、H3蛋白和mRNA表达显著升高,灯盏乙素干预可显著提高Aβ作用下细胞的存活率,显著降低H2A、H2B、H3 mRNA及蛋白表达。结论:灯盏乙素能调节Aβ毒性所致神经元中组蛋白H2A、H2B、H3的表达,可能通过改变AD表观遗传的信息异常来发挥预防和治疗作用。Objective:To investigate the effect of scutellarin(Scu)on the expressions of histone H2 A,H2 B and H3 in SH-SY5 Y cells treated with β-amyloid(Aβ),and to explore the possibility of the treatment of Alzheimer′s disease(AD)in epigenetic.Methods:SH-SY5 Y cells were divided into control group,Scu group,Aβ group and Aβ+Scu with different dose groups.Cell counting Kit-8(CCK-8)was used to detect cell survival rate.Cell apoptosis rate was determined by flow cytometry.The protein and mRNA expressions of H2 A,H2 B,H3 were examined by Western blot and real-time PCR,respectively.Results:Compared with the control group,the cells survival rate was decreased significantly,the apoptosis rate was increased significatly,and the protein and mRNA expressions of H2 A,H2 B,H3 were increased significantly in Aβ group.After treated with Scu,the cell surrival rate under the action of Aβ was significantly increased,the protein and mRNA expressions of H2 A,H2 B,and H3 were significantly decreased.Conclusion:Scu can regulate the expressions of histones H2 A,H2 B and H3 in neurons caused by Aβ toxicity,it is possible to play a preventive and therapeutic role by changing the information of AD epigenetic.
