电针关元对肾虚不孕大鼠性激素及其受体表达影响的实验研究

摘　　要：目的观察电针关元对肾虚不孕大鼠性激素及受体表达的影响,探讨电针关元治疗肾虚不孕的机理并评价疗效。方法对60只肾虚不孕模型SD大鼠进行研究,随机分为:正常组,模型组,关元组,非经非穴组,每组15只。用羟基尿灌胃15d获得肾虚不孕大鼠模型,在灌胃的同时各组予相应处理,第11天各组雌鼠与雄鼠按2∶1自由合笼4d受孕,第15天取出合笼的雄鼠,继续饲养,雌鼠于7d后无菌条件下处死,记录妊娠大鼠数。收集血清标本并测定血清促排卵素(FSH),黄体生成素(LH),雌二醇(E2),10%福尔马林固定子宫,并采用HE染色观察形态学改变。应用Western-blot法检测雌激素受体(ERα)和孕激素受体(PR)表达情况。结果电针关元可以有效改善肾虚排卵障碍大鼠的肾虚症状,促进子宫内膜和腺体发育,提高妊娠率,提高血清E2、LH、FSH含量并促进ERα及PR表达。结论电针关元可以有效改善肾虚不孕大鼠肾虚症状,并有效提高大鼠妊振率,其具体分子作用机制可能是通过调节子宫ERα和PR表达,促进子宫内膜和腺体发育,从而促进胚胎着床并提高受孕率。Objective To observe the effect of electroacupuncture at Guanyuan on the expression of sex hormones and receptors in rats with kidney-deficiency infertility,and explore its mechanism and evaluate its curative efficacy.Methods 60 SD rats with kidney-deficiency infertility were randomly divided into normal group,model group,Guanyuan group and non-meridian group,with 15 rats in each group.Kidney-deficiency infertility rats were induced by intragastric administration of Hydroxyurea for 15 days.On the 11th day,the female rats and the male rats were conceived in 2∶1 cage for 4 days.On the 15th day,the male rats were taken out of the cage,and then the female rats were sacrificed under aseptic condition after 7 days.Serum FSH,estradiol(E 2)and LH were measured by ELISA.And 10%formalin were used to fix the uterus.The morphological changes were observed by HE staining.The expression of estrogen receptor(ER alpha)and progesterone receptor(PR)was detected by Western blot.The number of embryos was recorded in pregnant rats.Results Electroacupuncture at Guanyuan could effectively improve the symptoms of kidney deficiency and promote the development of endometrium and gland in rats with ovulation disorder due to kidney deficiency.After treatment,the expressions of ER and PR in Guanyuan group were higher than those in model group.Conclusion Electroacupuncture at Guanyuan can effectively improve the symptoms of kidney-deficiency infertility and increase the rate of pregnancy.Its molecular mechanism may be to promote the implantation of the endometrium and glands by regulating the expression of ER-alpha and PR in the uterus,thereby promoting embryo implantation and increasing the pregnancy rate.
