摘 要:细菌性干枯病是发生在广东杧果产区杧果上的一种新病害,为明确该病的病原菌种类,通过稀释涂布法分离和纯化、致病性测定,并结合形态学、生理生化特性及16S rDNA序列分析,对病原菌的种类进行鉴定。结果表明,从杧果干枯病病样中分离获得的菌株NY01,革兰氏染色呈阴性,菌体杆状,最适培养温度28℃,最适pH值7.0~7.2。刺伤接种到健康杧果叶片,4 d后叶片呈现典型病斑。以菌株NY01的DNA为模板,扩增获得的16S rDNA片段,经BLAST分析,菌株NY01 16SrDNA序列与Sphingomonas sanguinis(登录号KT766073)的亲缘关系最近,同源性达到99.63%。经形态学、生理生化及16S rDNA序列分析将菌株NY01鉴定为血红鞘氨醇单胞菌S.sanguinis。血红鞘氨醇单胞菌能够引起杧果细菌性干枯病在国内属首次报道。The pathogen of a new bacterial disease dry rot was isolated in Guangdong province,and identified with dilution separation method,morphological characteristics and 16 S rDNA sequences analysis.The isolated strain was named NY01.It is gram-negative rod-shaped bacteria.The optimum culture temperature is 28℃,and the optimum pH value is 7.0 to 7.2.The NY01 strain was inoculated on mango leaves.The typical disease symptom appeared on the leaves at 4 days after inoculation with bacteria suspensions.Sequence of 16 S rDNA segments amplified from the NY01 shared 99.63%of homology with the sequence of Sphingomonas sanguinis(KT766073)from Genbank by BLAST analysis.Through morphology,physiology and biochemistry and 16 S rDNA sequence analysis,the strain NY01 was identified as S.sanguinis.It is the first report that S.sanguinis can cause bacterial dry rot on mango in China.
