摘 要:目的探究夹脊电针对ALS-SOD1^G93A转基因小鼠腰髓前角运动神经元β-catenin及GSK-3β表达的影响。方法选取18只ALS-SOD1^G93A转基因阳性雌性小鼠为实验对象,随机分为模型组、手针组、电针组,每组6只,另取6只同窝野生型雌性小鼠为阴性对照,小鼠8周龄时开始实验:阴性对照组不给予任何干预;模型组给予捆绑处理4周;手针组于双侧L1-2、L5-6夹脊穴给予普通针刺治疗4周;电针组于双侧L1-2、L5-6夹脊穴给予电针治疗4周。期间观察小鼠一般活动状况,并通过转棒实验评价小鼠协调性、力量和平衡能力。于16周取小鼠腰膨大L1-6腰髓,釆用免疫组化S-P法检测脊髓β-catenin、GSK-3β、p-GSK-3β的表达水平。结果与对照组相比,模型组转棒时间从12周开始逐渐减少,手针组、电针组从13周开始减少(P<0.05)。与模型组相比,手针组、电针组转棒时间下降趋势较为缓慢,差异有统计学意义(P<0.05).β-catenin、GSK-3β、p-GSK-3β主要在小鼠脊髓前角运动神经元细胞质中表达。与对照组相比,模型组小鼠脊髓前角β-catenin表达下降(P<0.05);与模型组相比,手针组、电针组β-catenin、β-GSK-30表达升高,p-GSK-3β/GSK-3β比值增加(P<0.05);与手针组比较,电针组β-catenin、p-GSK-3β表达升高,p-GSK-3β/GSK-3β比值升高(P<0.05)。结论夹脊电针通过抑制GSK-3β的活性,上调前角运动神经元β-catenin,对神经元细胞产生保护作用。Objective To explore the effect of Jiaji electro - acupuncture on the expression of GSK -3β and β- catenin in the lumbar anterior horn motor neurons of ALS - SODI^G93A transgenic mice. Methods A total of 18 ALS 一 SODI^G93A transgenic female mice were selected as experimental subjects, which were randomly divided into model group, hand 一 acupuncture group and electro - acupuncture group, with 6 mice in each group. Six wild - type female mice were taken as negative control. The experiment began when the age of the mice reached 8 weeks. No intervention was given in the negative control group. The model group was treated with bundling for 4 weeks;the hand acupuncture group was treated with ordinary acupuncture at bilateral L1 -2 and L5 -6 Jiaji points for 4 weeks;the electro - acupuncture group was treated with electro - acupuncture at bilateral L1 -2 and L5-6 Jiaji points for 4 weeks. During this period, the general activity of mice was observed, and the coordination , strength and balance ability of mice were evaluated by rod rotation test. Lumbar spinal cord of mice with lumbar enlargement was taken at 16 weeks. The expression levels of β- catenin, GSK -3β and ββ- GSK - 3β in lumbar spinal cord were detected by immunohistochemical S - P method. Results Compared with the control group, the rotating rod time of the model group decreased gradually from 12 weeks, while that of the hand - acupuncture group and the electro - acupuncture group decreased from 13 weeks ( P < 0. 05). Compared with the model group, the declining trend of rotating rod time in the hand - acupuncture group and the electro - acupuncture group was slower, and the difference was statistically significant ( P <0.05).β- catenin, GSK - 3β and β- GSK - 3β were mainly expressed in the cytoplasm of the motor neurons in the anterior horn of the spinal cord of mice.
