摘  要:目的观察冠心病(CHD)患者血清微小核糖核酸335(miR-335)、潜在转化生长因子结合蛋白2(LTBP-2)的表达,并探讨其意义。方法将102例CHD患者按疾病类型分为稳定型心绞痛(SAP)36例(SAP组)、不稳定型心绞痛(UAP)32例(UAP组)、急性心肌梗死(AMI)34例(AMI组),根据Gensini积分将102例CHD患者分为轻度病变组37例、中度病变组40例、重度病变组25例,另取冠状动脉(冠脉)造影诊断为非CHD患者40例为对照组。采用AU5800全自动生化分析仪检测空腹血糖、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平。采用实时荧光定量聚合酶链反应检测血清miR-335,采用酶联免疫吸附法检测血清LTBP-2水平。CHD患者病情严重程度与血清miR-335、LTBP-2的关系采用Pearson积矩相关性分析。结果对照组、SAP组、UAP组、AMI组收缩压、舒张压、空腹血糖、TG、TC、LDL-C、HDL-C比较差异均无统计学意义(P均>0.05)。对照组、SAP组、UAP组、AMI组血清miR-335水平呈逐渐下降趋势,而血清LTBP-2水平呈逐渐升高趋势,单因素方差分析结果显示,各组间血清miR-335、LTBP-2比较差异均有统计学意义(P均<0.05)。轻度病变组、中度病变组、重度病变组血清miR-335水平呈逐渐下降趋势,而血清LTBP-2水平呈逐渐升高趋势,单因素方差分析结果显示,各组间血清miR-335、LTBP-2差异均有统计学意义(P均<0.05)。Pearson相关分析结果显示,CHD患者病变严重程度与血清miR-335水平呈负相关(r=-0.673,P<0.05),而与LTBP-2水平呈正相关(r=0.705,P<0.05)。结论CHD患者血清miR-335呈低表达,而LTBP-2呈高表达;二者联合检测有助于CHD患者病情程度的评估。Objective To observe the expression of serum microribonucleic acid-335(miR-335) and latent transforming growth factor binding protein 2(LTBP-2) in patients with coronary heart disease(CHD), and to explore its significance.Methods Totally 102 patients with CHD were divided into 36 cases of stable angina pectoris(SAP)(SAP group), 32 cases of unstable angina pectoris(UAP)(UAP group), and 34 cases of acute myocardial infarction(AMI)(AMI group) according to the type of disease. According to Gensini score, 102 CHD patients were divided into the mild lesion group(37 cases), moderate lesion group(40 cases), and severe lesion group(25 cases). Another 40 patients with non-CHD diagnosed by coronary angiography were taken as the control group. The levels of fasting blood glucose, triglyceride(TG), total cholesterol(TC), low density lipoprotein cholesterol(LDL-C), and high density lipoprotein cholesterol(HDL-C) were measured by automatic biochemical analyzer. Real-time fluorescence quantitative polymerase chain reaction and enzyme-linked immunosorbent assay were used to detect the levels of serum miR-335 and LTBP-2, respectively. The relationships between severity of CHD and serum levels of miR-335 and LTBP-2 were analyzed by Preason correlation.Results There were no significant differences in systolic blood pressure, diastolic blood pressure, fasting blood glucose, TG, TC, LDL-C, or HDL-C between the control group, SAP group, UAP group, and AMI group(all P>0.05). The levels of serum miR-335 in the control group, SAP group, UAP group, and AMI group decreased gradually, while the level of serum LTBP-2 increased gradually. The result of one-way anova showed that there was significant difference in serum level of miR-335 and LTBP-2 between groups(all P<0.05). The serum level of miR-335 in the mild, moderate and severe lesion groups decreased gradually, while the serum level of LTBP-2 increased gradually, the result of one-way anova showed that there was significant difference in serum miR-335 and LTBP-2 between groups(all P<0.05
