摘  要:目的研究miR-29binhibitor基因修饰的骨髓间充质干细胞成骨分化及成软骨分化的潜能。方法2017年1月—2018年1月内蒙古医科大学选用新西兰大白兔2只,使用miR-29binhibitor慢病毒质粒转染兔骨髓间充质干细胞,将其基因修饰的骨髓间充质干细胞体外分别向成骨及成软骨诱导分化,采用碱性磷酸酶、茜素红、甲苯胺蓝、番红O、及Ⅰ、Ⅱ型胶原免疫组化染色进行检测,并与兔P3代骨髓间充质干细胞对比。结果Ⅰ型胶原免疫细胞化学染色实验组A(6.02±1.67)、实验组B(32.01±8.09)与对照组(0.33±0.52)比较,差异有统计学意义(P<0.05),实验组A与实验组B比较,差异有统计学意义。这表明miR-29binhibitor基因修饰的骨髓间充质干细胞成骨分化能力较P3代骨髓间充质干细胞弱(P<0.05);Ⅱ型胶原免疫细胞化学染色实验组A(7.33±2.16)、实验组B(6.83±1.94)与对照组(0.33±0.52)比较,差异有统计学意义(P<0.05),实验组A与实验组B比较,差异无统计学意义(P>0.05),成软骨分化能力两者相似。结论miR-29binhibitor基因修饰的骨髓间充质干细胞在诱导剂作用下具有向成骨细胞和成软骨细胞分化的潜能,为组织工程中关节软骨及软骨下骨损伤的修复提供思路。Objective To study the potential of osteogenic differentiation and chondrogenic differentiation of Bone Marrow Mesenchymal Stem Cells(BMSCs) modified by miR-29b inhibitor gene.Methods From January 2017 to January 2018,two New Zealand white rabbits were selected from Inner Mongolia Medical University to transfect rabbit bone marrow mesenchymal stem cells with miR-29b inhibitor lentivirus plasmid.Bone marrow mesenchymal stem cells (BMSCs) modified with BMSCs were differentiated into osteoblasts and chondrocytes in vitro.Alkaline phosphatase,alizarin red,toluidine blue,vanillin O,and type Ⅰ,Ⅱ collagen were used to detect the differentiation of bone marrow mesenchymal stem cells.And compared with rabbit P3 generation bone marrow mesenchymal stem cells.Results I type collagen immunocytochemical staining experimental group A:(6.02±1.67),experimental group B(32.01±8.09) compared with the control group (0.33±0.52),there was significant difference (P<0.05),experimental group A and experimental group B In comparison,the difference was statistically significant.This indicates that the osteogenic differentiation of bone marrow mesenchymal stem cells modified by miR-29b inhibitor gene is weaker than that of P3 bone marrow mesenchymal stem cells (P<0.05);type II collagen immunocytochemical staining experimental group A (7.33±2.16),experiment group B (6.83±1.94) compared with the control group (0.33±0.52),the difference was statistically significant (P<0.05).There was no significant difference between the experimental group A and the experimental group B (P>0.05).Similar.Conclusion The the BMSCs modified with miR-29b inhibitor gene have the potential to dif ferentiate into osteoblasts and chondroblasts under the action of an inducer,and provide ideas for the repair of articular cartilage and subchondral bone injury in tissue engineering.
