摘 要:目的 探讨miR-143在卵泡刺激素(FSH)介导的卵巢颗粒细胞孕酮生成中的作用。方法 实时定量RT-PCR检测FSH诱导原代颗粒细胞后miR-143的表达;重组腺病毒感染颗粒细胞进行miR-143的过表达;放射免疫法(RIA)测定孕酮的水平。结果 50ng/ml的FSH处理原代颗粒细胞24h后颗粒细胞分泌孕酮显著增加(P<0.001),建立了FSH刺激原代颗粒细胞分泌孕酮模型。miR-143在FSH处理颗粒细胞6、12h后表达没有显著变化,在24h后表达显著增加(P<0.01),在48h时表达量最高(P<0.001)。与感染对照病毒Ad-miRNC组相比,感染Ad-miR143组的细胞miR-143表达水平显著增加(P<0.01)。感染Ad-miR143后颗粒细胞与空白对照组和感染对照病毒组相比,孕酮分泌水平显著升高(P<0.01)。结论 miR-143可以促进FSH介导的卵巢颗粒细胞孕酮生成。Objective : To explore the role of miR-143 in FSH-mediated progesterone production in ovarian granulosa cells. Methods : The expression miR-143 in primary granulosa cell model induced by FSH was detected by real-time RT-PCR. The granulosa cells were infected with the recombinant gadenovirus,in order to over-express miR-143. The level of progesterone was determined by radioimmunoassay(RIA ). Results : After treated with 50 ng/ml FSH for 24 hours,the secretion of progesterone by granulosa cells was significantly increased( P <0.001),indicating that the differentiation model of primary granulosa cells was established. The expression of miR-143 was not significantly changed after 6 and 12 hours of FSH treatment,but it was significantly increased after 24 hours( P <0.01),and reached the highest level at 48 hours( P <0.01). Compared with the control virus Ad-miRNC group,the expression of miR-143 in the cells infected with Ad-miR143 group was significantly increased( P <0.01). Compared with the blank control group and the infected control group,the progesterone level secreted by granulosa cells infected with Ad-miR143 was significantly increased( P <0.01). Conclusions : miR-143 can promote FSH-mediated progesterone production in ovarian granulosa cells.
