摘 要:目的探讨右美托咪定诱导家兔心动过缓是否与窦房结缝隙连接基因蛋白Cx45和Cx31.9表达变化有关。方法将36只8周龄健康家兔分为3组,各12只,均通过耳缘静脉注射构建家兔心动过缓模型。巴比妥钠麻醉后,进行心电图、血压监测。A组持续注射生理盐水15 mL/kg;B组注射右美托咪定20μg/kg负荷量10 min,10μg/(kg·h)持续注射50 min;C组注射右美托咪定80μg/kg负荷量10 min,40μg/(kg·h)持续注射50 min。观察3组家兔平均动脉压(MAP)和血液流变(HR)变化情况,记录数据后解剖,取出家兔心脏,分离窦房结组织,通过免疫组化和荧光定量PCR检测Cx45和Cx31.9的表达水平。结果Cx45 mRNA表达水平,A组高于B组,B组高于C组(P<0.05);Cx31.9 mRNA表达水平,B组高于A组(P<0.05),C组与B组无明显差异(P>0.05);Cx45和Cx31.9的蛋白表达变化趋势与mRNA一致。结论右美托咪定诱导家兔心动过缓,导致窦房结缝隙连接基因蛋白Cx45和Cx31.9表达升高。Objective To investigate whether dexmedetomidine-induced bradycardia in rabbits is associated with the expression of sinoatrial node gap junction gene proteins Cx45 and Cx31.9.Methods Totally 36 8-week-old healthy rabbits were divided into three groups,12 rabbits in each group.The rabbit models with bradycardia were established by the marginal ear vein injection.After anesthesia with barbital sodium,electrocardiogram and blood pressure monitoring were performed.Group A was continuously injected with15 m L/kg normal saline,group B was injected with 20 μg/kg loading dose of dexmedetomidine for 10 min and continuous injection with 10 μg/(kg·h) for 50 min,and group C was injected with 80 μg/kg loading dose of dexmedetomidine for 10 min and continuous injection with 40 μg/(kg·h) for 50 min.The changes in mean arterial pressure(MAP) and hemorheology(HR) were observed in the three groups.After recording the data,the rabbit hearts were removed,and the sinoatrial node tissues were isolated.The expression levels of Cx45 and Cx31.9 were detected by immunohistochemistry and quantitative fluorescence PCR.Results The expression level of Cx45 in group A was higher than that in group B,and that in group B was higher than that in group C(P < 0.05).The expression level of Cx31.9 mRNA in group B was higher than that in group A(P < 0.05),but there was no significant difference between group C and group B(P > 0.05).The change trend of protein expression of Cx45 and Cx31.9 was consistent with that of mRNA.Conclusion Dexmedetomidine-induced bradycardia in rabbits can cause increased expression of sinoatrial node gap junction gene proteins Cx45 and Cx31.9.
