摘 要:目的本研究分析乌梅提取物对肾结石大鼠肾功能及中性粒细胞明胶酶相关脂质载运蛋白/单核细胞趋化因子蛋白1(NGAL/MCP-1)水平的影响,为相关药物研发及治疗提供参考。方法清洁级SD大鼠随机分为4组:对照组、模型组、乌梅提取物组和m TOR抑制剂雷帕霉素组。除对照组尾静脉注射生理盐水外,其余各组大鼠采用尾静脉注射纳米细菌复制肾结石模型,对照组和模型组给予羧甲基纤维素钠灌胃治疗,乌梅提取物组(20 mg/kg)和m TOR抑制剂雷帕霉素组(10mg/kg)大鼠给予对应药物治疗。分析各组大鼠尿液中24 h尿生化指标[尿酸(UA)和尿柠檬酸(CA)]及肾脏NGAL/MCP-1和m TOR蛋白表达变化。结果与对照组相比,模型组大鼠血清CA及UA水平明显升高(P<0.05),肾组织NGAL、MCP-1、AKT及m TOR蛋白的表达水平明显升高(P<0.05);与模型组比较,雷帕霉素组及乌梅提取物组大鼠血清CA及UA水平明显降低,肾组织NGAL、MCP-1、AKT及m TOR蛋白的表达水平明显降低(P<0.05)。结论乌梅提取物对肾结石大鼠肾功能及NGAL/MCP-1和AKT/m TOR水平的异常有明显的恢复作用。Objective The aim of this study was to analyze the effects of Wumei extract on renal function and NGAL/MCP-1 level in rats with renal calculi. Methods SD rats of clean grade were randomly divided into four groups: control group,model group,Wumei extract group and rapamycin group,an inhibitor of m TOR. In addition to normal saline injected into tail vein of control group,rats in other groups were injected nanobacteria into tail vein to replicate renal calculi model. The control group and model group were treated with sodium carboxymethylcellulose intragastrically. The rats in Wumei extract group( 20 mg/kg) and rapamycin inhibitor group(10mg/kg) were treated with corresponding drugs. The urinary biochemical indexes and urinary biochemical parameters at 24 hours before and after treatment were analyzed in urine and changes of NGAL/MCP-1 and m TOR protein expression in kidney. Results Compared with the control group,the levels of serum CA and UA in the model group were significantly higher( P<0.05). The expression levels of NGAL,MCP-1,AKT and m TOR protein in renal tissue were significantly increased( P<0.05). The above abnormalities were significantly restored in rapamycin group and Wumei extract group( P<0.05).Conclusion Wumei extract has obvious recovery effect on renal function and abnormal levels of NGAL/MCP-1 and AKT/m TOR in rats with renal calculi.
