摘 要:目的研究白藜芦醇和香青兰总黄酮对哮喘小鼠肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)水平的影响。方法 80只小鼠随机分白藜芦醇用药组(空白组、模型组、白藜芦醇组和地塞米松组,每组10只);和香兰总黄酮用药组(香兰总黄酮组,空白组模型组和地塞米松组,每组10只);分组后,对各组小鼠称重。构建哮喘小鼠模型,出现阳性反应后5周。白藜芦醇用药组:空白组和模型组生理盐水10μl/g灌胃,白藜芦醇组灌胃95 mg/kg白藜芦醇溶10μl/g,地塞米松组小鼠灌胃1 mg/kg地塞米松溶液10μl/g。香青兰总黄酮组:香青兰总黄酮组小鼠灌胃香青兰总黄酮溶液95 mg/kg,其余同白藜芦醇用药组。每天给药1次,连续给药30 d。实验后,对各组小鼠测定体重,采用酶联免疫吸附(ELISA)法检测小鼠支气管肺泡灌洗液(BALF)中TNF-α和IL-6的含量。结果白藜芦醇用药组和香兰总黄酮用药组小鼠BLAF中IL-6、TNF-α和体重比较,差异有统计学意义(P <0.05);进一步两两比较,与空白组相比,模型组小鼠的BALF中IL-6和TNF-α含量增多(P <0.05);与模型组相比,白藜芦醇组、香青兰总黄酮组和地塞米松组哮喘小鼠的BALF中IL-6和TNF-α降低(P <0.05),但白藜芦醇组哮喘小鼠BLAF中IL-6的抑制作用低于地塞米松组(P <0.05),而香青兰总黄酮组TNF-α含量高于地塞米松组(P <0.05);与模型组相比,地塞米松组、香青兰总黄酮组和白藜芦醇组小鼠体重下降(P <0.05)。结论白藜芦醇和香青兰总黄酮对哮喘小鼠炎性因子TNF-α和IL-6的释放具有一定抑制作用。Objective To study the effects of resveratrol and moldavica total flavone on tumor nerosis factor-α( TNF-α) and interleukin-6( IL)-6 in asthma mice. Methods 80 mice were randomly divided into resveratrol group( blank group,model group,resveratrol group and dexamethasone group,10 in each group);and moldavica total flavonoids group( vanilla total flavonoid group,and the rest were grouped the same as the first group,10 in each group);after grouping,mice of each group were weighed;5 weeks after the positive reaction developed,an asthma mouse model was constructed. In the resveratrol group,saline 10 μl/g was administered to the blank group and the model group;the resveratrol group was intragastrically administered with 95 mg/kg resveratrol 10 μl/g. The dexamethasone group were intragastrically administered with 1 mg/kg dexamethasone solution 10 μl/g;in the moldavica total flavonoids group,the moldavica total flavonoids group was intragastrically administered with moldavica total flavonoid solution 95 mg/kg,and the rest was the same as the resveratrol group. The drug was administered once a day for 30 days. After the experiment,the body weight of each group of mice was measured,and the contents of TNF-α and IL-6 in BALF of mice were detected by ELISA. Results The differences of levels of IL-6,TNF-α and body weight in the BLAF between the resveratrol group and the moldavica total flavonoids group were statistically significant( P < 0. 05). Further comparison showed compared with the blank group,the levels of IL-6 and TNF-α in the BALF of the mice in the model group increased;compared with the model group( P < 0. 05),the IL-6 and TNF-α showed compared with the blank( P < 0. 05),the total flavonoid group and the dexamethasone group decreased( P < 0. 05),but the inhibition of IL-6 in BLAF of asthma mice in resveratrol group was lower than that of dexamethasone group( P < 0. 05),and the content of TNF-α in moldavica total flavonoids group was higher than that of dexamethasone group( P < 0. 05).
