摘 要:目的:利用高脂饲料建立大鼠肥胖模型,通过对添加不同剂量反式脂肪酸(TFAs)喂养的大鼠脑及血液氧化损伤相关指标比较,探讨TFAs对肥胖大鼠脑和血液抗氧化系统的影响机制。方法:健康雄性SD大鼠40只,随机分为4组:对照组(CON组)、喂饲基础饲料、饮食诱导的肥胖组(DIO组),喂饲高脂饲料,1%及8%反式脂肪酸组(1%TFAs、8%TFAs组)喂饲添加1%TFAs及8%TFAs的高脂饲料。8周后,心脏取血后,处死动物,留取脑组织及血液以检测相关指标。结果:不同饲料喂养8周后,8%TFAs组大鼠脑丙二醛(MDA)含量明显高于CON组、DIO组与1%TFAs组(P<0.01);CON组、DIO组、1%TFAs及8%TFAs组大鼠脑还原型谷胱甘肽(GSH)含量依次显著下降,差异有统计学意义(P<0.01);1%TFAs组大鼠血清谷胱甘肽过氧化物酶(GSH-PX)活力低于DIO组、CON组(P<0.05);与CON组相比,8%TFAs组大鼠脑核因子E2相关因子2(Nrf2)mRNA表达水平显著上调(P<0.01);8%TFAs组血红素加氧酶-1(HO-1)蛋白表达水平显著高于CON组(P<0.05)、DIO组(P<0.05)及1%TFAs组(P<0.01)。结论:高含量TFAs摄入可引起肥胖大鼠血液和脑发生氧化损伤,上调氧化损伤相关基因和蛋白表达。Objective To observe the changes of oxidative damage indicators in the blood and brain of obese rats induced by high-fat diet with different doses of trans fatty acids(TFAs)and explore its mechanism of the effect.Method A total of 40 male rats were randomly divided into four groups(n=10),including control group(CON),diet-induced obesity group(DIO),1%TFAs group,8%TFAs group.These rats were fed with basal diet,high-fat diet,the animals were drew blood by cardiac,then anaesthetized with 1%chloral and killed.Brain tissue high fat diet with 1%TFAs and high fat diet with 8%TFAs respectively.At the end of 8th weeks,and serum was collected for related indexes detection.Result The content of MDA of 8%TFAs group in the brain was significantly higher than that of CON group,DIO group and 1%TFAs group(all P<0.01).The content of GSH of CON group,DIO group,1%TFAs and 8%TFAs group in the brain was significantly declined in turn(P<0.01).The GSH-PX activity of 1%TFAs group in serum was lower than that of the DIO group and the CON group(both P<0.05).Compared with CON group,the mRNA expression level of Nrf2genein 8%TFAs group was significantly up regulated(P<0.01).In addition,the expression level of HO-1 protein in 8%TFAs group was significantly elevated than that of CON(P<0.05),DIO(P<0.05)and 1%TFAs groups(P<0.01).Conclusion High concentration of TFAs contributes to the damage of antioxidant system of obese rats,which may be associated with the up-regulation of related genes and proteins expression.
