摘 要:[目的]研究亚香棒虫草多糖C-10体外抗肿瘤活性。[方法]以亚香棒虫草分离筛选的内生真菌为菌种,液体转化发酵产虫草多糖,经过分离提取得胞内和胞外多糖,并探讨其对3株癌细胞(肺腺癌A549细胞、乳腺癌MCF-7细胞、神经母癌SH-SY5Y细胞)的生长抑制效果。[结果]随着时间和浓度的增大,胞内多糖对癌细胞的抑制率逐渐增大,且差异较为明显,在培养72h的条件下,BN-H多糖(0.0966mg/mL)对肺腺癌A549细胞、乳腺癌MCF-7细胞、神经母癌SH-SY5Y细胞的抑制率分别为97%、71%、97%,比其他浓度的胞内外多糖抑制效果更好。[结论]BN-H(0.0966mg/mL)的亚香棒虫草多糖对肺腺癌、乳腺癌和神经母癌细胞具有极显著抑制作用(P<0.01),具有较强的时间和浓度依赖性,且胞内多糖对癌细胞的抑制效果好于胞外多糖。[Objective] The research aimed to study the anti-tumor activity of Cordyceps hawkesii polysaccharide C-10 in vitro.[Method]The endophytic fungi isolated from Cordyceps hawkesii were used as strains to produce cordyceps polysaccharides by liquid transformation.The intracellular and extracellular polysaccharides were isolated and extracted,and the inhibitory effects on the growth of three cancer cells (A549 cells,breast cancer MCF-7 cells and neuroblastoma cancer SH-SY5Y cells) were investigated.[Result]With the increase of time and concentration,the inhibition rate of intracellular polysaccharides on cancer cells was gradually increased,and the difference was obvious.Under the condition of raising 72h,the inhibitory rates of cancer A549 cells,breast cancer MCF-7 cells and neuroblastoma cancer SH-SY5Y cells were 97%,71% and 97% respectively,which were better than those of other concentrations of extracellular polysaccharides.[Conclusion]The polysaccharide of BN-H (0.096 6 mg/mL) can inhibit lung adenocarcinoma,breast cancer and neuroblastoma significantly ( P <0.01),and has a strong time and concentration dependent effect on lung adenocarcinoma,breast cancer and neuroblastoma ( P <0.01).The inhibitory effect of intracellular polysaccharides on cancer cells was better than that on extracellular polysaccharides.
