摘 要:目的了解黄芪、冬虫夏草对新生鼠肾积水病肾转化生长因子-β1(TGF-β1)和胰岛素样生长因子-1(IGF-1)表达的影响,探讨其对病肾损害有否保护作用。方法选用生后48h内的Wistar鼠96只,随机分为8组,各12只。随机选1组行假手术,作为对照组;余7组采用腰大肌压迫左输尿管上段制作动物模型,再任选1组做模型组(生理盐水1mL/kg/d),余6组随机分为黄芪组[低剂量1mL/kg/d+高剂量3mL/kg/d]、冬虫夏草组[低剂量0.5mL/kg/d+高剂量2mL/kg/d]、联合用药组[低剂量(黄芪1mL/kg/d+冬虫夏草0.5mL/kg/d)和高剂量(黄芪3mL/kg/d+冬虫夏草2mL/kg/d)],并按相应剂量定时皮下注射。饲养3周处死动物,切取不同组别的病肾组织,HE染色观察组织结构改变,免疫组化及RT-PCR检测病肾TGF-β1、IGF-1的蛋白及基因变化;收集肾盂尿液、ELISA法检测尿TGF-β1、IGF-1水平。结果对照组双肾大小、形态一致,肾小球、肾小管发育正常,肾组织内TGF-β1低表达、IGF-1高表达。模型组病肾较健肾大,肾盂扩张、积水,肾小球结构异常,肾小球鲍曼囊扩张、肾小管扩张;病肾组织TGF-β1在肾小管高表达、IGF-1低表达;病肾尿TGF-β1升高、IGF-1降低。黄芪、冬虫夏草组病肾虽仍大于健肾,但镜下肾小球、肾小管的损害较模型组明显减轻;病肾组织TGF-β1、IGF-1的蛋白及基因不同程度表达,尿TGF-β1、IGF-1水平介于对照组和模型组之间,与相应的对照组、模型组比较,差异均有统计学意义(P<0.05)。结论黄芪、冬虫夏草可能通过调控新生鼠肾积水病肾组织TGF-β1和IGF-1的表达,以阻止或减轻病肾的损害,从而起到保护效应。Objective To investigate influences of Astragalus and Cordyceps on transforming growth factor -β 1 (TGF-β1) and Insulin-like growth factor-1(IGF-1) of Hydronephrotic kidneys (HnK) in neonatal rat with partial ureter obstruction (PUO) and protection effect of Astragalus and Cordyceps in HnK in PUO. Methods 96 wistar rat which were born in 48 hours were randomly divided into 8 groups and there were 12 rats each group.And there were respectively sham operated as control group (CG) and made model group (MG) and experiment groups (EG).They were injected saline (0.9% NS 1.0mL .kg -1 .d -1 ) after operation in subcutaneous in CG.The experiment groups were made animal model of HnK by using psoas muscles compression left partial ureter.And to inject saline (0.9% NS 1.0mL .kg -1 .d -1 ) after operation in subcutaneous in MG and Astragalus(low dose 1.0 mL .kg -1 .d -1 + high dose 3.0 mL .kg -1 .d -1 )and Cordyceps (low dose0.5ml .kg -1 .d -1 + high dose 2.0ml .kg-1.d-1) in each respectively EG.To cut renal tissue of HnK in PUO and make examination specimen and collect urine samples.Pathological changes were conducted by HE staining and TGF-β 1 and IGF-1expression by immunohistochemistry and RT-PCR (polymerase chain reaction).Urine TGF-β 1 and IGF-1 were measured by ELISA (Enzyme-linked immuno-sorbent assay). Results There were equal volume and consistent shape of two kidneys by the naked eye in CG.And developing normally glomerulus and renal tubular and Bowman's capsule,and low TGF-β 1 and high IGF-1 expression in two kidney by microscope.The volume of HnK was enlarger gradually than that of health kidney in MG,and developing abnormally glomerulus and dilated Bowman's capsule and renal tubular,abundant TGF-β 1 and poor IGF-1 expression of renal tubular in HnK in PUO.The damage degree of glomerulus and renal tubular of Hydronephrotic kidneys was alleviativer obviously in each EG than that of HnK in MG although volume of HnK was also larger than that of health kidney in each EG.The TGF-β 1 expression of HnK was higher
