单核/巨噬细胞上调表达S100A8/A9在脓毒症中的作用研究

摘　　要：目的研究脓毒症中单核/巨噬细胞S100A8/A9表达水平的变化,初步探讨其与血清促炎细胞因子和炎症损伤的关系,为临床免疫调控提供理论依据。方法收集2012年1月至2012年12月在本院ICU病房治疗的21例脓毒症患者外周血标本,其中男12例,女9例。选择16例正常人外周血作为对照。采用流式细胞术检测各组外周血单核细胞上S100A8/A9的表达水平。结果流式细胞术分析发现,脓毒症患者外周血单核细胞上S100A8/A9的表达水平明显高于正常人,差异有统计学意义(P<0.001)。结论S100A8/A9在单核/巨噬细胞的上调表达可能是启动脓毒症早期炎症反应的关键步骤。Objective To examine the expression of S100A8/A9 in monocytes and macrophages in sepsis,and to assess its association with serum level of pro-inflammatory cytokines and inflammation damages. Methods The blood samples were obtained from 21 patients with sepsis,which were admitted to the Intensive Care Unit in our hospital during January 2012 to December 2012.The normal controlled group of 16 cases was set up as contrast.The expression of S100A8/A9 was determined by flow cytometry in peripheral blood monocytes. Results The expression of S100A8/A9 in monocytes were significantly higher in sepsis patients compared with the healthy controls ( P <0.001). Conclusion The up-regulated expression of S100A8/A9 in monocytes and macrophages may be a key step to initiate the inflammatory response in the early stage of sepsis.
