摘 要:目的建立UPLC法测定不同海拔和不同采收期乌梅Prunus mume(Sieb.) Sieb. et Zucc.中枸橼酸和苹果酸的含有量。方法乌梅80%甲醇提取物的分析采用ACQUITY HSS C18色谱柱(2.1 mm×100 mm,1.8μm);流动相乙腈-0.2%磷酸水,梯度洗脱;体积流量0.2 mL/min;检测波长210 nm;柱温35℃。结果枸橼酸、苹果酸分别在0.516~1.596、0.496~1.627μg/mL范围内线性关系良好(R2≥0.9996);平均加样回收率分别为100.36%、100.39%,RSD 1.43%、1.54%。结论该方法准确稳定,重复性好,可用于乌梅的质量控制。AIM To establish a UPLC method for the content determination of citric acid and malic acid in Prunus mume(Sieb.) Sieb. et Zucc. from different altitudes and different gathering periods.METHODS The analysis of 80% methanol extract of P. mume was performed on a 35 ℃ thermostatic ACQUITY HSS C18column(2.1 mm×100 mm,1.8 μm), with the mobile phase comprising of acetonitrile-0.2% phosphoric acid flowing at 0.2 mL/min in a gradient elution manner,and the detection wavelength was set at 210 nm.RESULTS Citric acid and malic acid showed good linear relationships within their own ranges(R2≥0.9996), whose average recoveries were 100.36%、100.39%,with the RSDs of 1.43%、1.54%,respectively.CONCLUSION This accurate,stable and reproducible method can be used for the quality control of P. mume.
