摘　　要：目的探讨蛋白酪氨酸磷酸酶1B(PTP1B)基因IVS6+G82A、Pro303Pro多态性与儿童肥胖的相关性。方法选取2015年2月至2018年2月在医院就诊的肥胖儿童303例(肥胖组),同时选取健康非肥胖儿童270例为对照组,采用聚合酶链反应-限制性片段长度多态性检测PTP1B基因IVS6+G82A、Pro303Pro多态性。结果 2组PTP1B基因IVS6+G82A基因型和等位基因分布无统计学意义(P>0.05);2组PTP1B基因Pro303Pro基因型和等位基因分布有统计学意义(P<0.05),其中肥胖组CT+TT基因型比例为32.66%;肥胖组IVS6+G82A各基因型患儿体重指数、腰围、腰臀比、体脂百分比等比较差异无统计学意义(P>0.05);肥胖组Pro303Pro基因型CC患儿体重指数、腰围、三酰甘油(TG)和低密度脂蛋白胆固醇(LDL-C)分别为(24.00±1.89)kg/m^2、(80.18±7.28)cm、(1.13±0.31)mmol/L和(2.02±0.61)mmol/L,明显低于CT和TT基因型患儿(P<0.05)。结论 PTP1B基因IVS6+G82A多态性与儿童肥胖无明显关系,而Pro303Pro多态性与儿童肥胖具有一定的相关性,值得进一步研究。Objective To explore the correlation between protein tyrosine phosphatase 1 B(PTP1 B) gene IVS6+G82 A and Pro303 Pro polymorphism and childhood obesity.Methods A total of 303 obesity children, who were treated in our hospital from February 2015 to February 2018, were enrolled as obesity group,at the same time, 270 healthy children were enrolled as control group. Polymerase chain reaction restriction fragment length polymorphism(PCR) was used to detect the polymorphisms of genes IVS6+G82 A and Pro303 Pro of the PTP1 B gene.Results There was no significant difference in the distribution of IVS6+G82 A genotype and allele between the two groups(P>0.05).However there was significant difference in the distribution of Pro303 Pro genotype and allele between two groups(P<0.05),in which the proportion of CT+TT genotype in obesity group was 32.66%. There were no significant differences in body mass index, waist circumference, waist hip ratio and body fat percentage, in different IVS6+G82 A genotypes in obesity group(P>0.05). The body mass index, waist circumference, triglyceride(TG) and low density lipoprotein cholesterol(LDLC) in obesity children with Pro303 Pro genotype CC were(24.00±1.89)kg/m^2,(80.18±7.28)cm,(1.13±0.31)mmol/L and(2.02±0.61)mmol/L, respectively, which were significantly lower than those of obesity children with CT and TT genotypes(P<0.05).Conclusion The polymorphism of PTP1 B gene IVS6+G82 A has no significant correlation with childhood obesity, but the polymorphism of genesPro303 Pr is correlated with childhood obesity, which is worth being researched further. The IVS6+G82 A polymorphism of PTP1 B gene is not associated with obesity in children,and Pro303 Pro polymorphism is associated with childhood obesity,is worth further study.
