摘 要:目的探讨TAZ基因在喉癌组织中的表达规律及其对喉癌Hep2细胞增殖的影响。方法免疫组化检测TAZ在32例喉癌及相应癌旁组织中的表达情况,实时PCR法检测其中TAZ mRNA的表达情况。喉癌Hep2细胞分为实验组和对照组,实验组转染真核表达载体pcDNA3.1-TAZ,对照组转染空载体,应用Western blot法及实时PCR法检测两组细胞中TAZ及EMT过程关键基因的表达水平;MTT法及细胞克隆形成实验测定两组细胞增殖活力。此外,转染pcDNA3.1-LATS1以激活Hippo通路检测TAZ在喉癌细胞中的作用是否依赖Hippo信号。结果喉癌组织中TAZ蛋白及mRNA表达显著高于癌旁组织(P<0.05);过表达TAZ后,Hep2细胞增殖活力明显高于对照组(P<0.05),其中EMT过程中上皮关键基因如E-cadherin下调(P<0.05),而间质细胞关键基因如Vimentin表达增加(P<0.05),同时活化Hippo后喉癌细胞EMT过程受抑制。结论TAZ在喉癌组织中高表达,TAZ过表达能够促进Hep2细胞增殖,且该过程依赖Hippo信号通路,提示Hippo-TAZ可能在喉癌发生、发展过程中起重要作用。Objective To investigate the expression of TAZ gene in laryngeal carcinoma and its effect on the proliferation of laryngeal carcinoma Hep2 cells.Methods Immunohistochemistry was used to detect the expression of TAZ in 32 of laryngeal carcinoma and adjacent tissues.The expression of TAZ mRNA was detected by real-time PCR.The Hep2 cells of laryngeal carcinoma were divided into experimental and control groups.The experimental group was transfected with eukaryotic expression vector pcDNA3.1-TAZ,and the control group was transfected into empty vector.Western blot and RT-PCR were used to detect the expression levels of key genes in TAZ and EMT in two groups of cells.MTT assay and cell clone formation assay were used to determine the proliferation activity of the two groups.In addition,transfection of pcDNA3.1-LATS1 to activate the Hippo pathway detected whether the role of TAZ in laryngeal cancer cells depends on Hippo signaling.Results The expression of TAZ and mRNA in laryngeal carcinoma tissues was significantly higher than that in adjacent tissues ( P <0.05).After over expression of TAZ,the proliferative activity of Hep2 cells was significantly higher than that of the control group ( P <0.05).The key genes such as E-cadherin were downregulated ( P <0.05) during EMT.The expression of key genes such as Vimentin in interstitial cells increased ( P <0.05).At the same time,the EMT process of laryngeal carcinoma cells was inhibited after Hippo activation.Conclusion TAZ is highly expressed in laryngeal carcinoma.TAZ overexpression can promote the proliferation of Hep2 cells.And the process depends on the Hippo signal pathway.It is suggested that Hippo-TAZ may play an important role in the occurrence and development of laryngeal carcinoma.
