摘  要:目的:观察桂枝甘草汤及其拆方对单个心室肌细胞膜电位相关参数的影响,分析桂枝甘草汤抗心律失常的电生理机制。方法:酶解法急性分离豚鼠心室肌细胞,分为空白组(灌胃生理盐水制备的血清孵育细胞)、甘草组(灌胃甘草30 g煎液)、桂枝组(灌胃桂枝60 g煎液)、甘草加桂枝组(灌胃甘草30 g煎液,桂枝60 g煎液)和桂枝甘草汤组(灌胃桂枝60 g,甘草30 g合煎液),应用全细胞膜片钳技术电流钳模式记录各组动作电位。结果:各组药物均引起细胞膜静息电位(resting membrane potential,RMP)减小,动作电位幅值(action potential amplitude,APA)降低,3相除极速率(V3)减慢,复极50%时程(APD50)、复极90%时程(APD90)和复极时程(APD)均延长,15%组药效显著(P<0.05)。结论:桂枝甘草汤含药血清引起心室肌细胞APA降低,APD50、APD90和APD均延长,各项指标变化趋势呈浓度依赖性,这可能是桂枝甘草汤具有抗心律失常作用的重要机制。Objective:To observe the effects of Guizhi Gancao Decoction and its decomposed formulas on the parameters related to membrane potential of single ventricular myocytes,and to analyze the electrophysiological mechanism of Guizhi Gancao Decoction against arrhythmia.Methods:Acute isolation of guinea pig ventricular myocytes by enzymatic hydrolysis was divided into blank group(serum incubating cells prepared by intragastric normal saline),licorice group(intragastric licorice 30 g decocted),Cinnamon Twig group(intragastric Cinnamon Twig 60 g decocted),licorice plus Cinnamon Twig group(intragastric licorice 30 g decocted plus Cinnamon Twig 60 g decocted and Cinnamon Twig Licorice Decoction group(intragastric Cinnamon Twig 60 g,Glycyrrhiza 30 g combined decoction).Action potentials of each group were recorded by operation current clamp mode.Results:Each group of drugs caused a decrease in the resting membrane potential(RMP),a decrease in the action potential amplitude(APA),a slowdown in the 3-phase depolarization rate(V3),and a repolarization 50% time course( APD50),repolarization 90% time course(APD90) and repolarization time course(APD) were prolonged,and 15% group was significantly effective(P<0.05).Conclusion:The drug serum contained in Guizhi Gancao Decoction caused a decrease in APA in ventricular myocytes,and APD50,APD90 and APD were prolonged.The trend of various indexes was concentration-dependent.This may be an important mechanism of Guizhi Gancao Decoction against arrhythmia.
