摘 要:目的:探讨绞股蓝七叶胆苷XVII对光老化小鼠血清中前列腺素2(prostaglandin 2,PGE2)、环氧酶2(cyclooxygenase 2,COX2)水平及皮肤组织中基质金属蛋白酶1(matrix matalloproteinase-1,MMP-1)表达的影响。方法:60只雄性小鼠随机分为空白对照组、UV模型组、七叶胆苷XVII高剂量组、七叶胆苷XVII中剂量组、七叶胆苷XVII低剂量组及阳性对照组,每组10只。各组给予相应药物13周后,分别用ELISA法测定蛋白水平,Western-blot法检测MMP-1水平。结果:与空白对照组比较,UV模型组血清中PGE2水平明显升高(P<0.01)。与UV模型组比较,绞股蓝七叶胆苷XVII各组血清PGE2水平显著下降(P<0.01)。与空白对照组比较,UV模型组血清中COX2水平明显升高(P<0.01)。与UV模型组比较,绞股蓝七叶胆苷XVII各组血清中COX2水平显著下降(P<0.01);与空白对照组比较,UV模型细胞中MMP-1表达明显升高(P<0.01)。与UV模型组比较,绞股蓝七叶胆苷XVII各组细胞中MMP-1表达显著下降(P<0.01)。结论:绞股蓝七叶胆苷XVII可能通过抑制PGE2、COX2水平及MMP-1表达防护皮肤光老化,可作为防治光老化的新型有效抑制剂。Objective:To investigate the effects of Gynostemma pentaphyllum aescinate XVII on serum prostaglandin 2(PGE2) and cyclooxygenase 2(COX2) levels and the expression of matrix metalloproteinase-1(MMP-1) in skin tissue of skin photoaging mice.Methods:60 male mice were randomly divided into blank control group,UV model group,high-dose group of aescinate XVII,middle-dose group of aescinate XVII,low-dose group of aescinate XVII and positive control group,with 10 mice in each group.After 13 weeks,the protein level was determined by ELISA and the MMP-1 level was detected by Western-blot.Results:Compared with the blank control group,the content of PGE2 in serum of the UV model group increased significantly(P<0.01).Compared with the UV model group,the serum PGE2 level in Gynostemma pentaphyllum XVII groups decreased significantly(P<0.01).Compared with the blank control group,the serum COX2 level in the UV model group was significantly higher(P<0.01).Compared with the UV model group,the serum COX2 level of Gynostemma pentaphyllum aescinate XVII groups decreased significantly(P<0.01).Compared with the blank control group,the expression of MMP-1 in UV model cells increased significantly(P<0.01).Compared with the UV model group,the expression of MMP-1 in the cells of Gynostemma pentaphyllum XVII groups decreased significantly(P<0.01).Conclusion:Gynostemma pentaphyllum aescinate may protect skin from skin photoaging by inhibiting the expression of prostaglandin 2,cyclooxygenase 2 and MMP-1.It can be used as a new effective inhibitor to prevent photoaging.
