摘  要:目的:研究仙鹤草药材不同药用部位的体外抗氧化、抗肿瘤、抗凝血活性,为生药不同部位的合理应用奠定实验基础。方法:采用HPLC色谱法建立仙鹤草不同药用部位的指纹图谱;采用MTT比色法研究仙鹤草不同药用部位对人肝癌细胞株HepG2细胞、人胃癌细胞株HGC-27细胞、人结肠癌细胞株Caco-2细胞增殖作用的影响;采用2,2’-联氮-(3-乙基苯并噻唑-6-磺酸)二铵盐(ABTS)法、1,1-二苯基-2-三硝基苯肼(DPPH)法检测仙鹤草不同药用部位的抗氧化能力;采用ELISA法测定仙鹤草不同药用部位对小鼠凝血酶原(PT)指标的影响。结果:仙鹤草不同药用部位的指纹图谱之间存在明显差异,根、茎、叶、花均显示不同程度抗肿瘤、抗氧化、抗凝血药效,其中茎对HepG2、HGC-27、Caco-2三种癌细胞株的增殖抑制作用强于根、叶、花;叶、花清除DPPH、ABTS自由基的能力强于根、茎;茎、叶降低小鼠血浆凝血酶原水平的能力强于根、花。结论:仙鹤草不同药用部位的抗氧化、抗肿瘤、抗凝血药效存在差异,为仙鹤草药材以药效为导向的不同药用部位的合理使用提供实验依据,为生药不同部位的精准应用奠定实验基础。Objective: To study the antioxidant, antitumor and anticoagulant activities of different medicinal parts of Herba Agrimoniae in vitro to lay an experimental foundation for rational application of different parts of crude drugs. Methods: HPLC method was applied to study the fingerprint of different medicinal parts of Herba Agrimoniae. The proliferation inhibition rate on HepG2, HGC-27 and Caco-2 was evaluated by MTT method. ABTS method and DPPH method were used to detect the antioxidant capacity of different medicinal parts of Herba Agrimoniae. ELISA method was used to detect the effect of different medical parts of Herba Agrimoniae on PT in rats. Results: The fingerprints of different parts of Herba Agrimoniae showed significant differences. Roots, stems, leaves and flowers showed different degrees of antitumor, anti- oxidation and anti- coagulation effects. Stems had stronger inhibitory effect on the proliferation of HepG2 cells, HGC-27 cells and Caco-2 cells than roots, leaves and flowers. The ability of leaves and flowers to scavenge DPPH and ABTS free radicals was stronger than that of roots and stems, and the ability of stems and leaves to reduce plasma prothrombin level in mice was stronger than that of roots and flowers. Conclusion: The antioxidant, antitumor and anticoagulant effects of different medicinal parts of Herba Agrimoniae are different, which provides experimental basis for the rational use of different medicinal parts of Herba Agrimoniae and lays experimental foundation for the accurate application of different parts of crude drugs.
