摘 要:目的研究赤灵芝的中枢镇静催眠作用,并初步探讨其作用机制。方法通过给予阈下剂量和阈剂量的戊巴比妥钠诱导小鼠睡眠(腹腔注射),记录其入睡情况、睡眠潜伏期和睡眠持续时间,用酶联反应法测定小鼠脑匀浆液中GABA、Glu、Gln、5-HT和5-HIAA含量。结果与蒸馏水组比较,阈下剂量的戊巴比妥钠可使赤灵芝组小鼠入睡率增加(P<0.05)。赤灵芝组小鼠脑内GABA、5-HT含量显著上升(P<0.01),Glu、Gln含量显著下降(P<0.01)。结论赤灵芝可能通过增加小鼠脑内GABA、5-HT含量,降低Glu、Gln含量,来发挥一定的镇静、催眠作用。Objective To study The sedative and hypnotic effects of Ganodermalucidum and explore its mechanism preliminarily. Methods Each group was given threshold dose and subthreshold dose pentobarbital sodium by intraperitoneal injection, and the sleep status, sleep latency,sleep duration of the mice were recorded. The contents of-aminobutyric acid(GABA), glutamate(Glu), glutamine(Gln), 5-hydroxytryptamine( 5-ht) and 5-hydroxyindoleacetic acid(5-HIAA) in rat brain homogenate were determined by enzyme-linked immunosorbent assay. Results Compared with the blank group, Ganodermalucidum group had an increased sleep rate after given subthreshold dose pentobarbital sodium(P<0.05). Compared with the blank group, brain GABA and 5-HT levels in Ganodermalucidum group increased(P<0.01),while the contents of Glu and Gln decreased(P<0.01).Conclusion Ganodermalucidum may play a calming and hypnotic role by increasing the content of GABA and 5-ht in the brain of mice and reducing the content of Glu and Gln.
