摘 要:采用CCK-8比色法检测大腹园蛛水溶性成分(WSFAV)对RAW264.7细胞的作用,并通过检测LPS刺激RAW264.7细胞后的IL-6和IL-1β的表达,证明WSFAV的体外抗炎效果。另外,利用小鼠急性腹腔炎症模型,进一步研究WSFAV的体内抑制炎症作用,并通过检测炎症细胞TLR-4的表达,初步证明WSFAV的抗炎机制。结果表明:WSFAV对细胞的活性无显著影响,但对炎症细胞表达IL-6和IL-1β具有明显抑制作用(P<0.05或0.01),并且WSFAV对小鼠急性腹腔炎症和炎症细胞的TLR-4的表达也有明显抑制作用。结果显示,WSFAV具有良好的抗炎作用,且其抗炎机制应该与TLR-4的表达相关。Cell Counting Kit-8(CCK-8)was used to determine the effects of the water soluble fraction of Araneus ventricousus(WSFAV)on RAW264.7 cells,and its anti-inflammation effects in vitro were proved by detecting the expression of IL-6 and IL-1βin inflammatory cells induced with lipopolysaccharide(LPS).The model of acute peritonitis in mice was further used to study the acute anti-inflammation effect of WSFAV in vivo,and the mechanism of the anti-inflammation had been preliminarily proved by detecting the expression of Toll-like receptor 4(TLR-4)in inflammatory cells.WSFAVs had no impact on the viability of RAW264.7 cells,whereas had evidently inhibitory effects on the expression of IL-6 and IL-1β(P<0.05 or 0.01).WSFAVs had significantly suppressive effects on the acute abdominal inflammation in mice and the expression of TLR-4 in inflammatory cell(P<0.05).WSFAV had significantly anti-inflammation effects by a mechanism which might be related to the expression of TLR-4.
