HPLC法同时测定黄柴安胃胶囊中盐酸小檗碱和黄芩素的含量

摘　　要：目的建立同时测定黄柴安胃胶囊中盐酸小檗碱和黄芩素含量的方法。方法采用高效液相色谱法,色谱条件:Athena C18柱,混合流动相:A为甲醇-乙腈(45∶55,V/V),B为乙腈-0.25 moL/L磷酸二氢钠水溶液(25∶75,V/V),梯度洗脱,流速:1.0 ml/min;0~25 min,检测波长:345 nm;26~60 min,检测波长:274 nm;柱温:32℃;进样量:10μl。结果盐酸小檗碱在1.235~96.246μg·ml-1范围内,黄芩素在3.265~205.248μg/ml范围内呈现良好的线性关系。加样回收率分别为100.40%(RSD=0.98%)和100.15%(RSD=1.02%)。结论该含量测定方法易操作、效率高、检测结果准确、稳定,可以用于黄柴安胃胶囊质量检测的方法。Objective A method for simultaneous determination of Berberine hydrochloride and Baicalin in Huangchai Anwei Capsule was established.Methods The chromatographic conditions were as follows: The chromatographic column:Athena C18,Mixed mobile phase: The mobile phase A:methanolacetonitrile(45∶55,V/V), The mobile phase B:acetonitrile-0.25 moL/L Sodium dihydrogen phosphate aqueous solution(25∶75,V/V), gradient elution was performed, the flow rate:1.0 ml·min-1, and 0 ~ 25 min,the detection wavelength:345 nm,26 ~ 60 min,the detection wavelength:274 nm and column temperature:32 ℃ and the sample size:10 μl. Results Berberine hydrochloride showed a good linear relationship within 1.235 ~ 96.246 μg/ml, and Baicalein within 3.265 ~ 205.248 μg/ml. The average recovery rate:100.40%(RSD = 0.98%) and 100.15%( RSD = 1.02%). Conclusion The method was simple, efficient, accurate and stable. It can be used for the quality determination of Huangchai Anwei Capsule.
