摘 要:目的探讨氟化钠(NaF)对大鼠骨组织碱性磷酸酶(alkaline phosphatase,ALP)和骨形态发生蛋白-2(bone morphogenetic protein-2,BMP-2)mRNA表达的影响。方法将96只8周龄SD雄性大鼠按体重随机分为4组,分别为对照(自来水)组和50、150、250 mg/L NaF染毒组,每组24只。采用自由饮水进行染毒,连续染毒24周。分别于染毒第8、16、24周时,每组选择8只大鼠,检测尿氟、血氟水平和氟斑牙及ALP、BMP-2 m RNA的表达水平。结果与对照组相比,不同浓度NaF染毒不同时间大鼠的尿氟、血氟浓度均升高,差异均有统计学意义(P<0.05);与第8周相比,不同浓度NaF染毒第16、24周大鼠的尿氟、血氟浓度均升高,差异均有统计学意义(P<0.05);且随着NaF染毒时间的延长和染毒浓度的升高,大鼠尿氟和血氟的浓度均呈上升趋势。与对照组相比,不同浓度NaF染毒不同时间大鼠的氟斑牙检出率均较高,除50 mg/L NaF染毒第8周外,差异均有统计学意义(P<0.05);随着染毒浓度的升高和染毒时间的延长,大鼠氟斑牙的检出率均呈上升趋势,损伤程度趋于严重。与对照组相比,不同浓度NaF染毒不同时间大鼠骨组织ALP m RNA的表达水平均较高,除50 mg/L NaF染毒第24周外,差异均有统计学意义(P<0.05);随着染毒浓度的升高,大鼠骨组织ALP m RNA的表达水平均呈上升趋势;与第8周比较,50、150 mg/L NaF染毒第16、24周大鼠骨组织ALP m RNA的表达水平均较低,而250 mg/L NaF染毒第16、24周大鼠骨组织ALP mRNA的表达水平均高,差异均有统计学意义(P<0.05);随着染毒时间的延长,50、150 mg/L NaF染毒大鼠骨组织ALP mRNA的表达水平均呈下降趋势,而250 mg/L NaF染毒大鼠骨组织ALP mRNA的表达水平呈上升趋势。与对照组比较,除50、150 mg/L NaF染毒第24周大鼠骨组织BMP-2 m RNA的表达水平较低(P<0.05)外,不同浓度NaF染毒不同时间大鼠骨组织BMP-2 m RNA的表达水平均较高,差异有统计学意义(P<0.05);随着染毒浓Objective To understand the effects of sodium fluoride exposure on m RNA expression of alkaline phosphatase(ALP)and bone morphogenetic protein-2(BMP-2) in bone tissue of rats. Methods A total of 96 male SD rats aged eight-week were randomly divided into four groups,24 rats per group,treated with sodium fluoride through drinking water at the doses of0,50,150 and 250 mg/L for 24 weeks,respectively. The levels of fluoride in urinary and serum samples,incidence rate of dental fluorosis,and the expressions of ALP mRNA and BMP-2 mRNA were detected by real-time PCR at the 8 th,16 th and24 th week of treatment. Results The fluoride concentrations in urine and serum showed significantly higher levels in 50-250 mg/L NaF groups with 8-24 weeks treatment compared with the control group(P<0.05),as well as in 16-week and 24-week NaF treatment groups compared with those in 8-week NaF treatment groups(P <0.05),and also showed increasing trend with the increase of treatment duration and doses.The detection rate of dental fluorosis showed significantly higher levels in groups with different NaF treatment doses and duration except 50 mg/L NaF group with 8-week treatment(P<0.05),and showed increasing trend with the increase of treatment duration and doses. The ALP m RNA expression in bone tissue revealed significantly higher levels in groups with different doses and duration of NaF treatment except 50 mg/L NaF group with 24-week treatment compared with the control group(P<0.05),and increased with the increasing exposure doses;Significantly lower expression levels of ALP mRNA were also found in 50 and 150 mg/L NaF groups with 16-week or 24-week treatment(P<0.05),whereas higher levels were found in 250 mg/L NaF group with 16-week or 24-week treatment(P<0.05),compared with those received 8-week treatment;The expression levels of ALP mRNA showed decreasing trend in 50 and 150 mg/L NaF groups,and increasing trend in 250 mg/L NaF groups,with the increase of treatment duration.Significantly lower expression levels of BMP-2 mRNA were found
